The genomic DNA analysis revealed that altarnative splice of exon 7 resulted the splice variant of KAI1. To analyse the role of splice KAI1 in motility and metastalic process of mouse colon adenocarcinoma (CT-26) cells, two stable cell lines that express wild-type (CT-26/wild KAI1) or splice variant of KAI1 (CT-26/splice KAU) were established. Immunoprecipitation study showed that the interaction of splice KAI1 with integrin α3β1 was more decreased than that of wild KAI1. Also, CT-26/splice KAI1 cells showed increased in vitro invasive potential and cell-extracellular matrix (ECM) adhesion as well as an increased in vivo tumorigenicity compared with those of CT-26/wild KAI1, These results indicate that the metastasis suppressor function of splice KAI1 was decreased compared with wild KAI1, and suggest that the functional difference of effects on cell motility, growth and metastasis between wild and splice KAI1 might be explained partially by structural differences of two KAI1 proteins, which are important in the interaction with other regulatory proteins.
