The cDNA contains an open reading frame of 3,378bp, encoding a protein with a predicted size of 126.8kDa. Sequence comparison of rTERT with known TERTs has uncovered four regions (v-I, v-II, v-III, and v-IV) conserved in the N-terminal halves of vertebrate TERT proteins and eight motifs (T, 1, 2, A, B. C, D, and E) conserved in the C-terminal halves of the known TERT protein. Real-time RT-PCR result revealed that an TERT expression pattern in rat was similar to that in mouse, rTERT mRNA was detected in normal adult tissues including testis, liver, spleen, brain, thymus, heart, kidney, prostate, bladder, muscle, and stomach. Western blot analysis revealed that rTERT protein was strongly detected in testis, liver, spleen, and brain. A comparison of the amino acid sequences of various TERTs and rTERT revealed high similarity to that of mouse TERT (86%) and low similarity to that of Tetrahynema TERT (39%). These data show that high conservation of structure and expression pattern between rTERT and mTERT suggests a conserved role for the protein in rat and mouse.
