In this thesis, have investigated the fabrications and characteristics of a polydimethylsiloxane-based microfluidic systems for a lab-on-a-chip. A low-cost, simple structured micropump, which is actuated by piezoelectric-discs, is fabricated using polydimethylsiloxane (PDMS). As a flow-rectifying element, diffusers are used instead of passive check valves. In order to evaluate the performance of a micropump, the deflection of a diaphragm and the flow rate of a pump are measured experimentally for various applying voltages. The deflection of a glass diaphragm is measured using an atomic force microscope (AFM). It increases linearly (up to 0.4㎛) with voltages when a square wave at 300 Hz is applied (up to 150 V) across a piezoelectric disc. The flow rate of a micropump increases with the applied voltage and decreases with the backpressure. The maximum flow rate and backpressure of a micropump are 32.9 ㎕/min and 173 Pa when 150 V square wave voltages are applied at 300 Hz. The micropump is numerically modeled based on the experimentally measured diaphragm deflection flow rate relationships, and the flow fields are analyzed using a computational fluid mechanics technique. The calculated flow rates are compared with the experimental measurements, and the results show that the performance of a micropump can be predicted accurately using a computational model. A normally-open thermopneumatic-actuated microvalve has been fabricated and its properties are investigated. The advantages of the proposed microvalve are the low-cost fabrication process and the transparent properties of the polydimethylsiloxane (PDMS) and indium tin oxide (ITO)-coated glass. The fabricated microvalves with in-channel configuration are easily integrated with other microfluidic devices on the same substrate. The fabrication process of thermopneumatic-actuated microvalves using PDMS is simple and its performance is suitable for a disposable lab-on-a-chip. The deflection of the PDMS membrane increases and the flow rate of the microchannel and microvalve decreases as the applied power to the ITO heater increases. The power to cut off flow is dependent on the membrane thickness and the applied inlet pressure but is independent of the channel width of the microvalves. The flow rate could be easily controlled by the switching function of the ITO heater and the closing and opening times were around 20 sec and 25 sec, respectively. A thermopneumatic-actuated polydimethylsiloxane(PDMS)-based micropump has been fabricated and its properties are characterized. Diffusers are used as flow-rectifying elements instead of passive check valves. The advantages of the proposed micropump are the low cost fabrication process and the transparent properties of the PDMS and indium tin oxide (ITO)-coated glass. We presented the PDMS micropump that is easily integrated with in-channel PDMS microvalves on the same substrate. The flow rate of the micropump increases linearly with increasing the applied pulse voltage to the ITO heater of which resistance is 6.54 ㏀ The peak flow rate is observed at the duty ratio of 10 % for the applied pulse voltage of 55 V at 6 Hz and the maximum flow rate of 78 nl/min is measured.
