Northern과 RT-PCR 분석 결과 AY84와 CA41의 발현이 저온 처리에 의해 증가된 반면 가뭄, 고 염분 그리고 낙엽산 처리에 의해서는 그 전사량이 감소되는 것으로 나타났다. Western 분석에 의해서도 AY84의 발현이 저온처리에 의해 유도됨이 확인되었다. Posttranscriptional regulations of gene expression in eukaryotes are of crucial importance for growth and development. The levels of regulation include pre-mRNA splicing, polyadenylation, mRNA transport, translation and stability. Regulation is mainly achieved either directly by RNA-binding proteins (RBPs) or indirectly, whereby RNA- binding proteins modulate the function of other regulatory factors. In recent years, a number of studies have demonstrated that the expression of RBP in plants increased in stress conditions, such as cold, drought, wounding, high salinity, and UV-irradiation, which imply that RBPs play roles in plant responses to environmental stresses. To understand the function of RBP and the mechanism of the RBP-mediated posttranscriptional regulation of genes in plants under various environmental stimuli, a number of target RBPs were identified among complete genome of Arabidopsis thaliana and the function of some of the selected genes was investigated. Both reverse transcription PCR (RT-PCR) and realtime RT-PCR were employed for an efficient screening of target genes induced upon various stress treatments including cold, drought, NaCl, and ABA. Among 14 potential genes tested, six RBP-encoding genes were highly induced by environmental stimuli, especially cold treatment, and two of these target genes, named AY84 and CA41, have been investigated in this study. The expression and function of the two genes were investigated by various biochemical and transgenic strategies. Northern and real-time RT-PCR analysis showed that the expressions of the AY84 and CA41 increased by cold treatment, whereas their transcript levels decreased by drought, salt, and ABA treatments. Western analysis also revealed that the expression of AY84 was induced by cold treatment. To obtain the recombinant RBP that could be used to search for the potential target RNA sequences in vitro, the RBP genes were sub-cloned into E. coli expression vector, and the expression conditions were optimized. To test the function of RBP in response to environmental stresses, the constructs for RNA interference and overexpression experiments for the AY84 have been generated. In addition to these in-house efforts to generate transgenic plants, the Arabidopsis T-DNA tagging mutant seeds with potential AY84 knockout mutation were obtained from the international consortium for Arabidopsis research, and the plants from the mutant seed were analyzed for the altered phenotype and different response to various environmental stresses. Results showed that the AY84 knockout mutant plant did not show any significant phenotype differences compared with the wild type plant. The present study have identified six potential RBPs in complete Arabidopsis genome and indicated the AY84 and CA41 as the RBPs highly induced by cold treatment, which provides a basis for further study to unravel the molecular mechanism of RBP-mediated posttranscriptional regulation of gene expression in stress response in plants.
