The N-terminal ends of titin, from adjacent sarcomere, overlap in the Z-line. The molecules span the I- and A- bands, and their C-terminal ends overlap in the M-line, thus forming a continuous filament system in myofibrils. We report here that the C-terminal domain of ankyrin-B interacts with the two most N-terminal immunoglobulin (Ig) domains of titin, ZIg1 and ZIg2, which are present at the Z-line. The interaction between ankyrin-B and ZIg1/2 titin were characterized using the yeast two-hybrid system, in in vitro pull-down assays and in experiments in COS -7 cells. Based on evidence showing that the C-terminus of ankyrin-B is necessary for the localization of titin at the Z-line, we propose that ankyrin-B may play a role in the organization of specific subdomains in the sarcolemma and the sarcoplasmic reticulum.
