(4) 생산된 단백질을 세포 밖으로 분비하므로 저렴하고 용이하게 분리 정제 할 수 있다. 따라서 식물 세포 배양 시스템은 높은 순도를 요하면서 소량으로 사용되는 고가의 의료용 단백질의 생산에 아주 적합한 시스템이다. 백혈병과 관련된 항암 보조치료제인 Human granulocyte colony stimulating factor (hG-CSF)를 생산하기 위하여 sucrose starvation inducible promoter 인 Ramy3D promoter와 3‘UTR을 이용하여 제작된 형질전환용 벡터에 도입하였고, particle bombardment mediated transformation을 이용하여 rice에 형질전환 하였으며, ELISA와 Northern blot 분석을 통하여 hG-CSF의 유전자 발현을 확인하였다. 시간대 별 ELISA 분석에서 hG-CSF 생산 및 축적률을 확인하였으며, 현탁 배양을 이용하여 생산된 hG-CSF의 최대 생산량은 2.5 ㎎/L 이였다. Bioassay를 통하여 biological activity 측정 결과, 식물세포 배양을 이용하여 hG-CSF가 bioactive form으로 생산되어짐을 확인하였다. 그러나 sucrose starvation 조건하에 벼의 현탁 세포에서 hG-CSF의 생산은 13일 후부터 protease의 분해에 의하여 단백질의 생산량이 급격하게 감소됨을 확인 하였으며 이를 억제하기 위하여 protease inhibitor 유전자를 발현하는 형질 전환된 벼의 현탁 세포와 함께 배양하므로 최적의 protein 생산 조건을 확립하였다. 이로써 형질 전환된 벼의 현탁 세포 배양 기술을 이용하여 면역학적으로 유용한 의료용 단백질 생산이 가능함을 확인하였다. Plants have provided humans with useful molecules for many centuries, but only in the past 20 years has it become possible to use plants for the production of specific heterologous proteins. The use of whole plants for the expression of recombinant proteins has received a great deal of attention because of advantages in economy, scalability and safety compared with traditional microbial and mammalian production system. However, production systems using whole plants lack several intrinsic benefits of cultured cells, including the precise control over growth conditions, batch-to-batch product consistency, a high level of containment and the ability to produce recombinant proteins in compliance with good manufacturing practice. Plant cell cultures combine the advantages of whole plant systems with those of microbial and animal cell cultures, and already have an established track record for the production of valuable therapeutic secondary metabolites. Plant cells are intrinsically safe, because (1) concerns about contamination of expressed proteins with human or animal pathogens (HIV, hepatitis viruses) or the co-purification of blood-borne pathogens and oncogenic sequences, are entirely avoided by using plants. (2) It is inexpensive system compared with an animal cell culture, because the medium consists of simple ingredients such as sucrose, inorganic salts, vitamins and hormones. (3) Post translational modification of the recombinant proteins occurs in the plant cells as in the mammalian cells. (4) Purification of the target protein is much easier than other expression systems because the plant cells can secrete foreign proteins into culture medium with relatively high concentration. Therefore, plant cell culture systems may be the most favorable means of producing small to medium quantities of high-priced, high-purity, specialty recombinant proteins, despite the fact that these systems are relatively new. Human granulocyte colony stimulating factor (hG-CSF), a human cytokine, was produced in transgenic rice cell suspension culture. Regulated expression and secretion of hG-CSF was achieved using the promoter, signal peptide, and terminator from a rice alpha-amylase gene Amy3D. The RAmy3D gene is expressed in response to sugar deprivation. The recombinant hG-CSF was secreted into the transgenic rice cell culture on sugar starved condition. Integration and expression of the hG-CSF gene were confirmed by genomic DNA PCR and Northern blot analysis, respectively. The amount of hG-CSF in culture medium on the sugar starved condition was determined by ELISA. The maximum amount of hG-CSF obtained in the rice cell culture medium with the RAmy 3D expression system was 2.5 ㎎/L. The recombinant hG-CSF produced through the rice cell culture system had biological activity in vitro in AML 193 cell proliferation assay, similar to the commercial E. coli-derived hG-CSF. The amount of recombinant hG-CSF protein in the medium increased until 13 days after induction, since then it decreased dramatically. Proteolytic enzymes existing in plant cell cultured media appear to be the major reason for the loss of secreted hG-CSF. The co-cultivation of transgenic rice cells expressing protease inhibitor appears to protect the secreted hG-CSF form the proteolytic degradation. The hG-CSF productivity could be increased 2-fold by using co-cultivation strategy. Therefore, the co-cultivation of the transgenic rice cell expressing protease inhibitor may be more than effective in stabilizing foreign proteins in plant cell suspension culture. Regulated expression and secretion of humanized F(ab')2 antibody was achieved using the promoter, signal peptide, and terminator from a rice alpha-amylase gene Amy3D. The RAmy3D gene is expressed in response to sugar deprivation. The recombinant humanized F(ab')2 antibody was secreted into the transgenic rice cell culture on sugar starved condition. Expression and secretion of assembled antibody was observed in transgenic rice suspension culture by Western blot analysis. Western blot analysis, ELISA, and FACS analyses confirmed that plant-derived hzAb is functional regarding its binding activity to the TAG 72. The hzAb expression was equivalent to 2% of the total secreted proteins. A purification step was developed to yield functional hzAb with purity greater than 95%. Furthermore, plant derived hzAb was as effective as animal derived hzAb in targeting to tumor of xenotransplanted LS 174T cells in nude mice. These data indicate that the hzAb derived from plant cell suspension culture uses a potential of pharmaceutical application for cancer therapy and diagnosis.
