This insecticide is widely used in top fruit, cotton and hops, and as a veterinary medicine for the treatment of ectoparasites in pigs, cattle, sheep, goats, and dogs. This chemical has been available in many countries since 1974, but amitraz poisoning in animals and humans has prominently increased in recent years because of the increased production and use volume. Its pharmacological and toxicological activities include monoamine oxidase inhibition, α2-adrenergic agonist activity, and prostaglandin synthesis inhibition. The present study was conducted to investigate the potential adverse effects of amitraz on pregnant dams and embryo-fetal development and to determine the potential reproductive and developmental toxicity of amitraz in Sprague-Dawley rats. In a embryo-fetal development study, the test chemical was administered by gavage to pregnant rats from days 1 to 19 of gestation at dose levels of 0, 3, 10, and 30 mg/kg/day. All dams were subjected to caesarean section on day 20 of gestation and their fetuses were examined for external, visceral and skeletal abnormalities. At 30 mg/kg, maternal toxicity included an increase in the incidence of abnormal clinical signs, a suppression in the body weight, and a decrease in the food intake. Developmental toxicity included an increase in the fetal deaths, a decrease in the litter size, and a reduction in the fetal body weight. In addition, an increase in the incidence of fetal external, visceral, and skeletal abnormalities was seen. At 10 mg/kg, slight maternal toxicity included a suppression in the body weight and a decrease in the food intake. In addition, minimal developmental toxicity including decreased fetal body weight, increased visceral and skeletal variations and increased fetal ossification delay was observed. There were no signs of either maternal toxicity or developmental toxicity at 3 mg/kg. The results show that amitraz administration during the entire period of pregnancy is embryotoxic and teratogenic at a maternally toxic dose (i.e., 30 mg/kg/day) and is minimally embryotoxic at a minimally maternally toxic dose (i.e., 10 mg/kg/day) in rats. In a reproductive and developmental toxicity study, the test chemical was administered via the drinking water containing 0, 40, 120, and 360 ppm to male rats from 2 weeks before mating to the end of 14-day mating period and to females from 2 weeks before mating, throughout mating, gestation and lactation up to weaning. During the test period, clinical signs, mortality, body weights, food and water consumption, reproductive organ weights, reproductive and littering findings, gross findings and histopathology were examined in the P generation. After birth of the F1 offspring, mortality, body weight change, physical signs of postnatal development and behavioral function were evaluated. When exposed offspring reached maturity, their reproductive capacity was assessed. Pregnant F1 females were euthanized on gestation day 13 and their F2 fetuses were examined. In the 360 ppm group, an increase in the incidence of abnormal clinical signs, a suppression in the body weight gain, a decrease in the food consumption, number of live pups at birth and number of live pups on postnatal day 4 and an increase in the post-implantation loss and a decrease in the relative weight of seminal vesicles were observed in the P generation. In addition, a growth retardation by suppression in body weight gain, a decrease in the grip strength, an increase in the peri- and post-implantation loss and a decrease in the number of live embryos on GD13 were observed in the F1 generation. In the 120 ppm group, an increase in the incidence of abnormal clinical signs, a suppression in the body weight gain, a decrease in the food consumption were observed in the P generation. A suppression in the body weight gain and a decrease in the grip strength were also observed in the F1 generation. There were no signs of either reproductive toxicity or developmental toxicity at 40 ppm. In conclusion, administration of amitraz to Sprague-Dawley rats caused various types of reproductive and developmental toxicities including an increase in the embryo-fetal deaths, a decrease in the litter size, a reduction in the fetal body weight, an increase in the incidence of fetal abnormalities, a decrease in the number of live pups and a suppression in the physical development. In the present experimental conditions, the no-observed-adverse-effect level (NOAEL) of amitraz by oral gavage is considered to be 3 mg/kg/day for both dams and embryo-fetal development and the reproductive and developmental NOAEL by drinking water was estimated to be 40 ppm (i.e., approximately 6 mg/kg/day).
