한우 송아지 설사증은 송아지 질병중 폐사율이 70%에 달해 경제적 피해를 가장 많이 주는 질병이다. 이 질병의 원인은 크게 바이러스성, 세균성, 원충성이라고 알려져 있다. 하지만 국내에서 한우 송아지 설사증 원인체에 대한 구체적 연구가 미흡한 실정이다. 따라서 한우 송아지 설사의 다발 원인체들에 대한 분리 및 특성을 규명하기 위하여 본 연구를 수행하였다. 먼저 한우 송아지에서 설사를 일으키는 바이러스성 원인체들을 검출하기 위해 A형을 비롯 B형과 C형 소 로타바이러스, 소 코로나바이러스, 소 바이러스성 설사 바이러스, 소 토로바이러스, 소 사포바이러스, 소 노로바이러스 등에 대한 특이적인 각각의 프라이머를 만들어, 92개 농가에서 생후 70일령 이하의 어린 송아지에서 채취한 설사분변 153개를 대상으로 RT-PCR을 수행하였다. 그 결과 81개 농가(88.0%)의 132개 설사분변(86.3%)에서 1가지 또는 2가지 이상의 바이러스가 검출되었으며 A형 소 로타바이러스의 경우 92개 농가 중 54개 농가(58.6%)에서 검출되어 가장 많은 분리율을 나타내었다. 그 외에 분변 내 소 코로나바이러스는 25개 농가(27.2%)에서, 소 바이러스성 설사 바이러스는 24개 농가(26.1%), 소 사포바이러스는 22개 농가(23.6%), 소 노로바이러스는 15개 농가(16.3%), 소 토로바이러스는 11개 농가(11.9%), C형 로타바이러스는 10개 농가(10.9%), B형 로타바이러스는 6개 농가(5.9%)에서 각각 검출되었다. 이들 바이러스의 감염양상을 보면, 단독감염이 36개 농가(44.5%)에서, 2중감염이 14개 농장(17.3%), 3중감염이 21개 농장(25.9%), 4중감염이 9개 농장(11.1%), 5중감염이 1개 농장(1.2%)에서 조사되었다. 이 결과로 볼 때 전남지방에서 발생하는 한우 송아지 설사는 주로 70일령 이하에서 발생되고, 장염 유발성 바이러스들이 주원인이었음을 확인 할 수 있었다. 또한 상당히 많은 설사가 발생한 송아지들은 다종의 바이러스들에 장내 혼합감염되어 있어서, 백신에 의한 예방도 어렵거니와 일단 설사증 발생시 치료 자체가 어렵다는 것을 알 수 있었다. 앞으로 이러한 바이러스들에 대한 병리학적 기전과 분자역학적인 연구는 물론 백신개발을 위한 더 많은 연구가 필요하다고 사료된다. 특히, 우리나라에서 A형 소 로타바이러스는 한우 산업에 큰 경제적 손실을 일으키는 원인체로 알려져 있지만 단지 바이러스를 분리하고 역학을 밝히는 제한적인 연구만 되어왔다. 따라서 국내에서 유행하고 있는 A형 로타바이러스를 분리하고 동정하기 위하여, A형 로타바이러스에 특이적인 프라이머를 이용하여 RT-PCR을 수행한 결과 양성으로 판정된 54개 농가에서 채취한 87개 설사분변을 fetal rhesus monkey kidney (MA104) 세포에 접종하여 바이러스 분리를 시도하였다. 그 결과 MA104 세포에서 세포변성 효과를 보이는 69주의 A형 소 로타바이러스가 분리(79.3%) 되었다. 또한 각각의 분리주들이 감염된 MA104 세포주를 대상으로 FITC conjugated hyperimmune gnotobiotic pig anti-BRVA serum을 이용한 면역형광항체법을 적용한 결과 RT-PCR과 같은 결과를 보였다. 실제로 소 설사분변내 A형 소 로타바이러스의 대규모 분리 및 동정에 대한 연구는 이 연구가 국내 처음이며, 이 결과는 향후 국내 A형 소 로타바이러스 설사증의 기병론 및 분자특성 규명과 백신개발 등에 유용한 기초자료로 활용될 것으로 판단된다. 한편 한우 송아지 설사의 중요한 원인체로 알려진 대장균에 대한 병원체로서의 역할은 그 동안 자세한 연구가 국내에서 수행되지 않았다. 일반적으로 설사를 유발하는 병원성 대장균은 현재 병원기전에 따라 장관침입성 대장균(enteroinvasive E. coli, EIEC), 장관독소원성 대장균(enterotoxigenic E. coli, ETEC), 장관출혈성 대장균(enterohemorrhagic E. coli, EHEC) 또는 vero 독소생성 대장균(verotoxin-producing E. coli, VTEC), 장관병원성 대장균(enteropathogenic E. coli, EPEC) 및 장관응집성 대장균(enteroaggregative E. coli, EAggEC) 등으로 분류되고 있다. 설사를 유발하는 E. coli의 병인기전과 관련된 독성인자와 이를 암호화하는 유전자들은 shiga toxin (stx), enterohemolysin (hlyA), intestinal adherence factor인 intimin (eaeA), lipopolysaccharides (rfbE) 등이 알려져 있다. Stx는 shiga 독소생성 E. coli (STEC)와 EHEC의 병원성을 지배하는 중요한 유전자이며 이는 Shiga toxin 1 (stx1)과 Shiga toxin 2 (stx2)로 나눌 수 있다. EHEC의 병원성 유발인자로는 stx 뿐만 아니라 intimin, enterohemolysin, lipopolysaccharide 등이 관여한다. 소는 STEC의 주요 보균동물로 알려져 있으며 이는 송아지 설사의 발병요인이 될 수 있다. 그래서 이 연구에서는 104개 농장의 255개의 한우 송아지 설사분변을 대상으로 STEC를 분리하고, stx 유전자형과 hlyA, rfbE 및 eaeA 유전자를 조사하였으며, 또한, 분리균주의 항생제 감수성 검사를 실시하였다. 검사된 송아지 설사분변에서 DNA를 분리하여 stx 공통 프라이머(STXc)를 이용하여 PCR을 실시한 결과 26개(10.2%)의 분변에서 518 bp의 특이 band가 증폭됨을 관찰할 수 있었다. 또한 26개의 STEC 분리주의 독소형을 알아보기 위해 stx1 및 stx2에 대한 프라이머를 이용하여 PCR을 실시한 결과, stx1에 대한 특이 양성반응인 130 bp의 특이 band가 총 18주에서 확인되었다. 그리고 stx2의 특이 양성반응인 378 bp의 특이 band가 총 10주에서 검출되었다. 그리고 stx1과 stx2가 동시에 관찰된 균주는 총 4주이었고 stx1과 stx2 어느 것도 갖지 않는 균주는 2주였다. 총 26주의 STEC 분리주에 대하여 병원성 유전자 보유상태를 조사한 결과, enterohemolysin인 hylA에 양성 반응(1,551 bp)을 나타내는 균주는 총 7주 (26%)였으며, eaeA (240 bp)에 양성반응을 보인 균주는 총 6주(23%)였다. 이들중 5주는 hlyA와 eaeA 유전자를 동시에 갖고 있었다. 또한 STEC 26주 중 1주만이 E. coli O157의 병원성 유전자인 rfbE에 특이적인 프라이머에 대한 양성 반응 (986 bp)을 보였다. 이들 분리 균주를 항혈청(O157:H7)을 이용하여 슬라이드 및 시험관 응집반응을 실시한 결과 E. coli O157:H7으로 판명되었다. 104개 농장 255개의 한우 송아지 설사분변에서 분리 동정한 STEC 26주에 대하여 amikacin외 11종의 항균제 감수성 검사를 실시한 결과 26주의 STEC 모두 penicillin에 대해 내성을 나타냈으며 이들 내성균 중 단제 내성균은 6주(23.1%)이었고 20주(76.9%)가 다제 내성균이었다. 다제 내성균 중 6제 내성균이 5주(19.2%)로 가장 많았고 3제 내성균은 하나도 검출되지 않았다. 그리고 tetracycline에 19주(73.1%), streptomycin에 18주(69.2%), kanamycin에 16주(61.6%), ampicillin에 15주(57.7%), cephalothin에 13주(50%), trimethoprim/sulfamethoxazole에 9주(34.6%), chloramphenicol에 7주(26.9%), gentamycin에 3주(11.5%), colistin에 1주(3.8%)가 각각 내성을 나타내었다. Amikacin에는 26주 모두가 감수성이 있는 것으로 나타났으며 gentamycin (88.5%), chloramphenicol (73.1%), trimethoprim/sulfamethoxazole (65.4%), amoxicillin-clavulanic acid (57.7%), colistin (50%)에 대하여는 중등도 이상의 감수성을 나타내었다. 본 실험에서 국내 한우 송아지 설사분변에서 분리한 STEC의 독소형과 유전자 성상을 규명할 수 있었으며 항생제 내성 정도를 파악할 수 있다는데 큰 의의가 있었다. STEC는 여러 축종에서 광범위하게 분포되어 있으며 공중보건학적으로도 중요한 인수공통전염병체로도 인정되고 있기 때문에 다양한 축종에서 계절 및 지역별로 분리하여 분자역학적 연구 등이 앞으로 수행되어야 할 것으로 사료된다.

To evaluate the enteric viruses involved in calf diarrhea, RT-PCR with primer pairs specific to each groups A, B and C bovine rotaviruses (BRV), bovine coronavirus (BCoV), bovine viral diarrhea virus (BVDV), bovine torovirus (BToV), bovine sapovirus (BSaV), and bovine norovirus (BNoV), respectively, was performed with 153 diarrheic calf fecal samples from 92 farms. Group A BRV was predominantly detected (54/92 farms) in the diarrheic fecal samples. BCoV (25/92), BVDV (24/92), BSaV (22/92), BNoV (15/92), BToV (11/92), group C BRV (10/92) and group B BRV (6/92) in order were also detected in the diarrheic fecal samples. Of calf farms with diarrhea, 132 fecal samples from 81 farms were positive for at least one of each virus. Among calf farms positive for each enteric virus, 36 farms were infected with only one virus of them. From these results, calf diarrhea in Korea was caused mainly by enteric viruses and considerable numbers of it was concurrently infected, leading the difficulty of vaccine or medical treatments. Further studies need to establish the pathogenicity, molecular epidemiology and vaccine development of these viruses with the massive diarrheic fecal samples. Although bovine group A rotavirus (BRV A) is known to cause huge economic losses to cattle industry in Korea, only limited studies including virus isolation and characterization have been reported. In order to isolate and characterize, virus isolation with fetal rhesus monkey kidney (MA104) cell lines and virus characterization by RT-PCR assay, targeting a 1062 bp fragment of the VP7 gene of BRV A, and immunofluorescence (IF) test with FITC conjugated hyperimmune gnotobiotic pig anti-BRV A serum were performed with 87 fecal suspensions of calves with diarrhea from 54 herds during 2004-2005 in Korea Jeonnam area. Among these diarrheic fecal samples, 69 BRV A isolates (79.3%) causing cytopathic effect on the MA104 cells were isolated. By RT-PCR assay, expected band was amplified with the culture supernatant of all BRV A isolates, indicating BRV A were successfully isolated. In addition, IF test revealed all isolates caused positive reaction in the cytoplasm of MA104 cells, supporting the above mentioned result. This is the first large scale study to isolate and characterize BRV A in calves with diarrhea in Korea. In order to decide what kinds of BRV A are currently popular and then develop BRV A vaccines, the further studies need to analysis the molecular characterization of these BRV A isolates, particularly VP7 and VP4 genes which induce virus neutralization together. Shiga-toxin (Stx) producing Escherichia coli (STEC) causes various clinical signs in animal and human. In this study, 255 fecal samples from Korean Native Calves showing diarrhea were collected from cattle farms in Jeonnam province during the period from January 2005 to July 2005. Twenty six STEC (10%) were isolated from 255 fecal samples by PCR. The isolates displayed three different stx combinations (stx1 [14 of 26], stx1 and stx2 [4 of 26], stx2 [6 of 26]). The isolates were further studied for virulence associated genes and antimicrobial resistance to define the virulence properties. Intimin (eaeA) gene, enterohemolysin (hlyA), lipopolysaccharide (rfbE) virulence genes were detected in 6 (23%), 7 (26%), 1 (3.8%) of the isolates, respectively, by PCR. One isolate possessing rfbE gene was typed as E. coli O157:H7 by agglutination test with O and H antiserum. All 26 isolates showed susceptibility to amikacin (100%) and the majority of isolates showed high susceptibility to gentamycin (88.5%) and chloramphenicol (73.1%). But all isolates were resistant to penicillin. These results might provide the basic knowledge to establish strategies for the treatment and prevention of enteric disease in Korean Native Calves.

In order to isolate and characterize, virus isolation with fetal rhesus monkey kidney (MA104) cell lines and virus characterization by RT-PCR assay, targeting a 1062 bp fragment of the VP7 gene of BRV A, and immunofluorescence (IF) test with FITC conjugated hyperimmune gnotobiotic pig anti-BRV A serum were performed with 87 fecal suspensions of calves with diarrhea from 54 herds during 2004-2005 in Korea Jeonnam area. Among these diarrheic fecal samples, 69 BRV A isolates (79.3%) causing cytopathic effect on the MA104 cells were isolated. By RT-PCR assay, expected band was amplified with the culture supernatant of all BRV A isolates, indicating BRV A were successfully isolated. In addition, IF test revealed all isolates caused positive reaction in the cytoplasm of MA104 cells, supporting the above mentioned result. This is the first large scale study to isolate and characterize BRV A in calves with diarrhea in Korea. In order to decide what kinds of BRV A are currently popular and then develop BRV A vaccines, the further studies need to analysis the molecular characterization of these BRV A isolates, particularly VP7 and VP4 genes which induce virus neutralization together. Shiga-toxin (Stx) producing Escherichia coli (STEC) causes various clinical signs in animal and human. In this study, 255 fecal samples from Korean Native Calves showing diarrhea were collected from cattle farms in Jeonnam province during the period from January 2005 to July 2005. Twenty six STEC (10%) were isolated from 255 fecal samples by PCR. The isolates displayed three different stx combinations (stx1 [14 of 26], stx1 and stx2 [4 of 26], stx2 [6 of 26]). The isolates were further studied for virulence associated genes and antimicrobial resistance to define the virulence properties. Intimin (eaeA) gene, enterohemolysin (hlyA), lipopolysaccharide (rfbE) virulence genes were detected in 6 (23%), 7 (26%), 1 (3.8%) of the isolates, respectively, by PCR. One isolate possessing rfbE gene was typed as E. coli O157:H7 by agglutination test with O and H antiserum. All 26 isolates showed susceptibility to amikacin (100%) and the majority of isolates showed high susceptibility to gentamycin (88.5%) and chloramphenicol (73.1%). But all isolates were resistant to penicillin. These results might provide the basic knowledge to establish strategies for the treatment and prevention of enteric disease in Korean Native Calves.
