본 연구에서는 한국산 참당귀(Angelica gigas Nakai)의 유효성분을 추출하기 위한 추출조건과 용매조건을 설정하고 새롭게 순도 높은 decursin 분리방법을 재정립 하였다. 1. 참당귀(Angelica gigas Nakai)의 일반성분은 수분 11.0%, 조단백질 13.5%, 회분 6.4% 및 탄수화물 66.5%로 나타났다. 또한 총 식이섬유 함량은 32.1%였으며, 그 중 불용성 식이섬유는 23.8%였으며, 수용성 식이섬유는8.3%였다. 2. 가용성 고형분은 95% EtOH 농도에서 28.7%로 가장 낮았으며, 50% EtOH 농도에서 38.2%로 가장 높게 나타났다. 총페놀 함량은 각각 extraction group별로 199.4 ppm, 297.8 ppm, 717.0 ppm, 400.3 ppm 및440.5 ppm으로 50% EtOH 농도에서 총페놀 함량이 가장 높았다. 3. 참당귀(Angelica gigas Nakai)의 함량은 분석방법중 이동상(mobile phase)의 조성에 따라 retention time이 각각 13.0분과 32.0분으로 나타났으며, decursin과 구조이성체인 decursinol angelate가 분리 되는 것을 확인하였다. EtOH의 농도가 증가할수록 decursin 함량이 증가하였으며, 50% EtOH 이상에서 decursin 함량을 90%이상 용출할 수 있었다. 따라서 대량생산에 필요한 효율적인 추출방법으로는 50% EtOH 이상에서 2시간씩 2회 추출이 가장 효율적이다는 결과를 얻을 수 있었다. 4. 당귀의 coumarin계 성분을 분리하여 융점 확인 111.3℃, 열적반응, 분자량 328.15(C_(19)H_(20)O_(5)), HPLC 정량, ^(1)H, ^(13)C-NMR 확인한 결과 decursin 구조이성체인 decursinol angelate가 제거가 된, decursin을 분리한 것을 확인 하였다. Decursin의 분리방법을 재정립함으로써 당귀를 이용한 제품 개발시, 품질관리 분석지표를 쉽게 설정할 수 있을 것으로 사료된다.

Also, it was analyzed to provide basic data for food materialization, processing and the usefulness of Angelica gigas Nakai as functional food materials. The contents of moisture, crude protein, crude fat, crede ash, carbohydrates and total dietary fiber of Angelica gigas Nakai were 11.0, 13.5, 2.6, 6.4, 66.5 and 32.1%, respectively. There were Five different solvent types for extraction process were used, i.e., distilled water, 25% EtOH, 50% EtOH, 75% EtOH and 95% EtOH. The results of total yield and total phenol in Angelica gigas Nakai with distilled water, 25% EtOH, 50% EtOH, 75% EtOH and 95% EtOH were 30.1, 31.3, 38.2, 35.8 and 28.7%, respectively and 199.4, 297.8, 717.0, 400.3 and 440.5 ppm, respectively. This study was progressed in two ways. The high performance liquid chromatography(HPLC) conditions(Method Ⅰ) were as follows; column: CAPCELL PAK C18, mobile phase: 70% methanol, detecter: UV 280 nm. The HPLC conditions(Method Ⅱ) were as follows; column: CAPCELL PAK C18, mobile phase: 52% acetonitrile, detecter: UV 280 nm. According to the analytical results, contents(%) of decursin with Method Ⅰ were higher than that of decursin with Method Ⅱ. The decursin was isolated and purified from fraction by Prep.-HPLC. A major of Angelica gigas Nakai, the decursin was identified by differential scanning calorimeter(DSC), mass spectrophotomter(MS), thermo gravimetric(TG), ^(1)H and ^(13)C-NMR analysis. The results of that, based on these studies, Angelica gigas Nakai may be developed into a potentially useful food materials.
