PCR법에 의한 진단은 Perkinsus atlanticus genomic DNA (Accession number, AJ238400)의 5S RNA와 V. tapetis genomic DNA (Accession number, AY129278)의 16S RNA에서 조합한 primer sets를 이용하여 현장조사를 하였으며, 그 결과를 종래의 진단방법과 비교하였다. V. tapetis 접종군의 폐사율은 고농도에서는 67.5%, 저농도에서는 7.5%에 불과하였으며, BRD의 특징적인 증상인 패각 내면의 chonchiolin 침착에 의한 갈색반점도 관찰되지 않았다. V. tapetis를 인위 감염시킨 바지락의 혈청 라이소자임 활성은 균 접종 1일 후부터 현저하게 감소하였으며, 혈구의 탐식율과 탐식지수는 시간이 경과할수록 현저히 감소하였으며 평균 탐식지수는 1.7이었다. 한편 균 접종 후 7일 까지 과립구의 비율은 감소하고 무과립구는 증가하는 경향이었다. V. tapetis의 PCR산물은 V. tapetis와 유사성이 높은 10종의 Vibrio균을 사용하여 특이성을 검정한 결과 오직 V. tapetis에 대해서만 414 bp의 특이밴드가 확인되었다. 2004년 4월부터 7월까지 태안과 고창 지역에 분포하고 있는 동죽, 바지락, 굴 및 피뿔고둥의 아가미에서 DNA를 추출하여 두 병원체를 탐지하기 위한 PCR 반응을 시킨 결과 Perkinsus sp.와 V. tapetis가 검출되었다. 2지역의 평균 감염률을 보면 Perkinsus sp.는 바지락이 95.0%, 굴은 10.0%, 동죽은 62.5%, 피뿔고둥은 46.9%로 나타났고, V. tapetis에서는 굴이 23.1%, 동죽이 9.4% 검출되었으며 바지락과 피뿔고둥에서는 검출되지 않았다. 2004년 인천 지역에서 발과 외투막이 수축되며 대량 폐사한 바지락의 조직 도말표본과 병리조직표본에 의한 검사한 결과 미잠입 개체는 100%, 잠입개체는 80%의 감염이 확인되었다. Perkinsus sp. and Vibrio tapetis infections of Manila clam (Ruditapes philippinarum) were investigated with some conventional methods such as Hemacolor stain, histopathological observations and FTM assay for Perkinsus sp. and cultivation on agar plates for V. tapetis from December 2001 to september 2003. PCR assay for their detection were developed and compared with the conventional methods. Numbers of viable cells, Vibrio species and V. tapetis, the causative organism of brown ring disease (BRD), in seawater, sediment and various tissues of Manila clam were counted on agar plates. A selective medium, marine agar (Difco) with 1% mannitol (w/v) and 1% bromthymol blue was applied for the isolation of V. tapetis. V. tapetis form bluish green colonies with halo because the bacteria can't utilize mannitol in the medium for their energy source. Pathogenicity of V. tapetis and dominant Vibrio species from seawater and Manila clam tissues were examined by artificially injecting them to Manila clam. Resistance of Manila clam against V. tapetis (NCIMB 13622) was also evaluated by checking activity of lysozyme in hemolymph, phagocytic activity of hemocytes and change of hemocytes composition. PCR assay was established by primer sets, which were partial sequences of 5S RNA of Perkinsus atlanticus genomic DNA (Accession number, AJ238400) and 16S RNA of V. tapetis genomic DNA (Accession number, AY129278) and compared with existing methods using for the detection of Perkinsus sp. and V. tapetis. V. tapetis was not isolated even though application of the selective agar plates for the bacterium. But Perkinsus sp. was detected in all surfacing and burrowing Manila clams. In conclusion, Perkinsus sp. and V. tapetis were proven to be widespread in culturing beds for Manila clams in the west coastal area of Korea. Histopathological observation was the most effective in investigation of Perkinsus sp. infection, which wasn't different from the results using by PCR assay. V. tapetis was not isolated by bacterial incubation on agar plates but detected by PCR assay. PCR assay to detect V. tapetis was proven to be effective.
