Secondly, the purpose of study in an effort to improve in vitro spermatogenesis by potentiating the interactions between developing germ cells, somatic cells, and the extracellular matrix (ECM), the efficiency of the germ cell-somatic cell co-culture in a 3-dimensional (3D) collagen gel matrix was examined. Finally, the purpose of this study was to estimate the efficacy of 3-D culture in a collagen gel matrix in supporting germ cell survival in long-term in vitro culture and resumption of meiosis and spermatid differentiation of human immature spermatogenic cells isolated from non-obstructive azoospermic men. Testicular cell was preparation from postnatal 18 day mice and Rat for Sertoli cell or whole testicular cell culture into the embedded collagen gel matrix. Collagen solution make up from rat tail. Dissociated cells were re-constructed in a collagen gel matrix and then cultured for up to 9 or 22 day in modified DMEM/F12 medium at 32°C with 5% CO2 in air. During culture, morphogenesis of collagen gel matrix with testicular cells was performed with H/E staining. After culture, the differentiation ratios of cultured cells were confirmed by immunohistochemitery, Realtime PCR, FACS scaning with haploid cell specific maker. After the few days of incubation, collagen gel matrices were contracted and firm testicular cell complex were formed. Based on immunocytochemistry, a haploid population of cells appeared in vitro that was not in 18-day-old mice testis. Viability of the cells cultured by monolayer method or in a collagen gel matrix was 55%, 75% respectively. Collagen gel matrix culture method was observation indicated that 75% of the TP2 antibody stained cells developed from seeding cells. And matrigel added collagen gel matrix was showed 85% of the TP2 antibody stained cells from seeding cells. In contrast, Monolayer culture method showed that only 20% of cultured germ cells developed to round spermatids. The biosynthetic and proteolytic behavior of mouse Sertoli cells cultured in crosslinked collagen gel matrix is different from that in monolayers or in floating collagen gels and more similar to that previously described in attached collagen gels. The novel culture system developed in this study is the first to promote activity, differentiation of cultured cells to presumptive haploid cells in vitro spermatogenesis. Compared to the conventional monolayer culture method, the system appeared to be superior. The clinical application of 3-dimensional culture in a collagen gel matrix provides a suitable means by which spermatocytes could be induced to differentiate into presumptive spermatids in vitro. In addition, plasma FSH level could be a good indicator for the differentiability of cultured spermatogenic cells obtained from spermatocyte-arrested patients. This work characterizes a novel and efficacious culture system that takes advantage of the unique properties of the extracellular matrix, a 3D microenvironment, and the presence of in vitro spermatogenesis.
