To investigate influence of the viral thymdine kinase (vTK) of herpes simplex virus (HSV) on intracellular uptake and anabolism of antiviral drugs and the role of vTK on latent establishment and reactivation in cell culture model, I examined with various acyclovir (ACV)-resistant laboratory isolates. TK-deficient mutants were classified into 2 groups, TK-partial (TKp) and TK-negative (TK-). Thymidine (Thd) uptake was highly dependent on viral TK activity of each virus and continued until the end of virus replication cycle. The TK-positive strain, F showed the highest, the TKp strains moderate although the levels varied among them and the TK- strains and mock little uptake. Influence of the vTK was much less in ACV than ganciclovir (GCV) and penciclovir (PCV). Thd incorporation could occur regardless of viral DNA replication. Inhibition of Thd incorporation by ACV, GCV or PCV did not decrease Thd uptake at all. Even notable increase by ACV was observed. There were only few differences in anabolism pattern among viral strains in each cell line. In FTK143B with vTK activity the difference of uptake levels of ACV, GCV and PCV among viral strains became minimized. The vTK expressed in cells as fusion proteins with fluorescent proteins by gene-transfection found mainly in the nucleus, but the vTK in virus-infected cells has been considered being mainly in the cytoplasm. I have performed intracellular localization of the vTK in virus-infected cells using immunofluorescence confocal microscopy. Regardless of viral strains or cell types, the vTK were observed mainly in the nucleus. They were detected in the cell nucleus from 2 h p.i. to the end of viral growth in one-step growth experiments. Apart from virus amplification imaging the localization of the vTK could be a good tool to confirm cell susceptibility to virus and to check homogeneity of cell population.HSV latency is readily established by both HSV-1 strain F and TK-deficient mutants in the presence of drugs in cell culture models, primary mouse trigeminal ganglion nerve cells (mTG) and rat differentiated PC-12 cells. HSV reactivation from latent infection analyzes with various inducer in cell systems. Although progress has been made, the cellular and viral functions responsible for the establishment, maintenance and reactivation of HSV latency are not well understood. Studying HSV infection in the isolated TK-deficient mutants in vitro may provide information regarding the regulation of the complex interactions between cellular and viral functions during HSV latency and reactivation.
