In this study carbon source-enhanced (fumarate) in situ biological treatment of stepwise reduction of nitrate to nitrite, nitrous oxide, and molecular nitrogen in a nitrate-contaminated groundwater by adapting an in situ well-to-well circulation process (WWCP) was estimated. Molecular methods to assess suspended microbial community dynamic of denitrifying microorganisms and compare the results with the field data obtained was also adapted. The field method consists of the extraction of the groundwater from an extraction well (EW) which is the most up-gradient well, and the controlled injection of a prepared test solution and the groundwater mixture into an aquifer through an injection well (IW). The injected test solution consists of groundwater containing a nonreactive tracer (bromide) and one or more biologically reactive solutes (fumarate and nitrate). PCR amplification of 16S rDNA genes and denaturing gradient gel electrophoresis (DGGE) were used to monitor changes in the microbial community. A WWCP tracer test and WWCP biodegradation tests were conducted in sequence. The tracer test was conducted to evaluate the mobility of solutes [bromide (a conservative tracer) and nitrate (an electron acceptor)] used in a subsequent test. The breakthrough curves for all solutes of monitoring well 1 (MW1) and monitoring well 2 (MW2) placed up-gradient and down-gradient to IW, respectively, were similar indicating the apparent conservative transport of the solutes. 20-200% denitrification fumarate injection tests (DFIT) where the amounts of fumarate needed to achieve 20-200% denitrification of nitrate injected and complete consumption of background DO of 3 mg/L were added, were conducted in sequence in the WWCP biodegradation tests using a continuous injection method. However, the bioclogging of IW screens occurred and it resulted in overflow of IW, probably due to the excess proliferation of heterotrophs around well screens. To minimize the bioclogging problem, a pulse injection method was adapted. WWCP adapting pulse injection with up-gradient extraction system reduced the level of nitrate concentration (as N) from 30 mg/L to below the detection limit, and maintained the concentration less than 1 mg N/L for nitrate, less than 0.4 mg N/L for nitrite, and nearly zero for nitrous oxide. The ranges of nitrate and nitrite concentration at the down-gradient of the system met the requirements, a maximum contaminant level (MCL) for nitrate (10 mg NO3--N/L) and nitrite (1 mg NO2--N/L) in drinking water set by US Environmental Protection Agency. Before the biostimulation of indigenous microorganisms, the dominant DGGE bands obtained by PCR were affiliated with γ – Proteobacteria consisting of Acinetobacter sp. and Pseudomonas fluorescens. However, as the biological denitrification proceeded, the dominant patterns of DGGE bands changed, and they were affiliated with Azoarcus denitrificans Td-3 and Flavobacterium xanthum. Azoarcus denitrificans Td-3 is known to completely reduce nitrate to nitrogen gas, suggesting that the denitrifying bacteria probably play an important role in in situ complete denitrification of nitrate. Pulse injection WWCP adapting biological denitrification is a useful means for remediating a nitrate-contaminated aquifer.
