Con A를 처리하여 세포들을 활성화를 시킨 후 8시간이 지났을 때 IL-2 mRNA의 생성은 가장 활발히 일어났고, IL-2R$\alpha$ mRNA의 수는 활성화 후 80시간이 지날 때까지 계속 증가하였다. 임상적 치료에서 면역억제제로 널리 쓰이는 cyclosporin A는 매우 낮은 농도에서도 IL-2 뿐만 아니라, IL-2R$\alpha$ 유전자의 발현도 강하게 억제하였다. 발암물질로 잘 알려져있는 AAF와 IQ 역시 두 유전자의 전사를 대조군에 비해 30\% 이하로 억제하는 결과를 나타내었다. 실험에 사용된 다른 면역억제제들 중에서 aflatoxin $B_1$, cyclophosphamide, TCDD는 IL-2 mRNA 발현을 억제하지 않았으나, DMBA는 생쥐의 간세포에 의해 활성화가 된 후에 IL-2 유전자의 전사를 억제하는 결과를 보였다.

In this study, the effects of various kinds of immunosuppressive agents on IL-2 and IL-2R$\alpha$ mRNA expression were analyzed in mouse splenocytes using competitive RT-PCR method. The accuracy of this PCR-based analysis method, developed in this study, was verified by the linearity between the amounts of input template and that of the resulting PCR product. After activation with Con A ($2 \mu g/ml$), IL-2 mRNA levels peaked at 8 hr, while IL-2R$\alpha$ mRNA transcripts increased gradually until 80 hr. Cyclosporin A, a well-known immunosuppressive drug, markedly inhibited IL-2R$\alpha$ expression, as well as IL-2, at very low concentration (10 ng/ml). AAF and IQ also reduced the mRNA expression of both, IL-2 and IL-2R$\alpha$, to less than 30\% of induced level. In the analysis of TCDD, no significant decrease in IL-2 mRNA transcript was detected. Among other chemicals (aflatoxin $B_1$, cyclophosphamide, DMBA) which need metabolic activation to exert their immunosuppressive effects, only DMBA showed inhibitory effects on IL-2 mRNA transcription after coculture with mouse hepatocytes.
