Cytokine messenger RNA (mRNA) was measured using reverse transcriptase polymerase chain reaction (RT-PCR) and cytokine protein production by enzyme linked immunosorbent assay (ELISA). A549 cells were exposed to 10 to 500 ㎍/㎖ of suspension of PM10 for 24 hours. Each was compared with those in the control non-exposed cells. The mRNA levels of interleukin (IL)-8, IL-Iβ, granulocyte macrophage colony stimulating factor (GM-CSF) were increased after exposed to PM10 collected during Asian dust period, compared to those in non-dusty period and the control, in a dose dependent manner. Cytokine production in supernatants of A549 cells incubated with 10 to 500 ㎍/㎖ of PM10 suspension of Asian dust period increased in a dose dependent manner for IL-8 and IL-1β, not in the control and non-dusty period. In conclusion, A549 cells exposed to PM10 collected during Asian dust period produce proinflammatory cytokines more than those during non-dusty period. These cytokines will enhancing the local inflammatory response in the airways and could also contribute to the systemic component of this inflammatory process.
