Menadione (이하 MD)은 활성산소종 (reactive oxygen species; ROSs) 가운데superoxide anion (O2-)을 유발하는 물질로 알려져 있다. KNU5377 균주는 다양한 비우호적인 환경 (adverse conditions) 하에서도 뛰어난 스트레스 내성 (stresstolerance)을 가지고 있기 때문에, 먼저 MD에 유도된 산화적 스트레스 (oxidative stress)에 대한 adaptive stress response (ASR)에 초점을 맞추었다. 연구는 직접적으로 120 μM의MD 농도 하에서 1시간동안 처리한 샘플 (전 처리하지 않은 경우; MD-treated cellswithout pre-adaptation)과 6 μM의 MD 농도 하에서 1시간 동안 전처리 후 120 μM의농도로 처리한 샘플 (전 처리한 경우; pre-adapted cells)로 나누어서 수행하였다. 먼저 스트레스 하에서 세포 내의 hydroperoxide, carbonyl content, 그리고 malondialdehyde level은 전 처리한 경우에 상당히 낮았다. DNA microarray를 이용한 transcriptional analysis에 있어, 전 처리하지 않은 경우 에는 detect된 약 6,000 여개의 유전자 가운데 782개의 유전자가 2배 이상 유도 되었고, 반면에 전 처리한 경우에는 1110여개의 유전자가 과발현 되었다. Saccharomyces cerevisiae, potential yeast species used in many fields of industrial fermentation, exhibits significant differences in bread, beer, and wine fermentation resulting from the genomic DNA. To survey the genetic diversity of S. cerevisiae KNU5377 strain, which presents the high fermentation capacity at strain levels, we performed various PCR profiling techniques. Among five S. cerevisiae strains analyzed, genetic differences of KNU5377 were well elucidated by PCR typing of pulse-field gel electrophoresis (PFGE), δ sequences, PCR differentiation using intron splice site primers, random amplified polymorphic DNA (RAPD) analysis with M13 primer, Sau-PCR, and microsatellite analysis, but not by restriction fragment length polymorphism (RFLP) analysis of internal transcribed spacers (ITS). Notably, minisatellite analysis and the nucleotide sequences of the AGA1 gene (YNR044w), the coding a-agglutinin anchorage subunit, proved to be highly polymorphic in KNU5377. The ORF (2519 bp) of AGA1, which contained two blocks of tandem repeat units as serine (Ser) and/or threonine (Thr)-rich block, was longer than that (2178 bp) of S. cerevisiae S288C, the positive control. The current results showed that genetic patterns of KNU5377 were strongly different from the other S. cerevisiae strains. We suggest that KNU5377 might have the different molecular mechanisms involved in the adaptive evolution of yeast traits by the specificities of the molecular basis of the physiological properties such as the generation of evolutionary novelties resulting from significant genetic polymorphism. Menadione (MD), a simplest synthetic quinone used with reactive oxygen species (ROSs) generating agents, causes damages to S. cerevisiae KNU5377. To understand antioxidant defense mechanisms under adaptive stress response, we have performed biochemical analyses and the genome-wide gene expression analysis after exposure to 120 μM of MD with and without adaptation. KNU5377 could survive at higher concentration of MD, 680 μM for 1 h, comparing with S. cerevisiae W303, ATCC24858 as reference strains. Glutathione peroxidase (GPX2), thioredoxin peroxidase 2 (TSA2), and thioredoxin reductase (TRR1) were up-regulated over 10-fold after treatment of 120 μM. Superoxide dismutase (SOD1), catalase (CTA1), glucose-6-phosphate dehydrogenase (ZWF1), glutathione peroxidase (GPX2, HYR1), glutathione reductase (GLR1), thioredoxin peroxidase (AHP1), and thioredoxin 2 (TRX2) were also strongly induced after adaptation. Additionally, transcriptional analysis of those genes was well corresponded with up-regulated protein expression and enzyme activity. Antioxidant defense mechanisms containing SOD1, ZWF and TRX2 maybe orchestrate by the different levels at the same time and function to produce effective defense systems. We also studied the transcriptional regulation using DNA microarray technology to adapt mandione (MD)-induced oxidative stress in a yeast S. cerevisiae KNU5377. Of ~6200 ORFs investigated, approximately 6000 genes were detected. In the MD-treated cells without pre-adaptation, 782 genes showed a > 2 fold higher mRNA level, whereas in the pre-adapted cells up-regulation of 1110 genes identified. Of the up-regulated genes in both cells, approximately 35% belonged to uncharacterized genes (hypothetical ORFs). In particular, the KNU5377 strain represented induction of trehalose -synthesis and/or -degradation genes (TSL1, TPS2, NTH1 and ATH1), genes (FAA1 and CAT2) involved in peroxisome functions, genes encoding reactive oxygen species (ROS)-scavenging systems such as superoxide dismutase (SOD1) and thioredoxin peroxidase (TSA1, AHP1, TSA2, DOT5) and glucose-6-phosphate dehydrogenase (ZWF1) in the both cells. Expression changes of the most genes encoding heat shock proteins (Hsps) such as Hsp104, Hsp90, and Hsp30 were stimulated although the expression intensities were not same. However, gene expression of HSP60 was not corresponded between mRNA level and protein level. As best showed by genome-wide analysis of gene expression, adaptive response to MD stress showed a more strong transcriptional stimulation of the genes involved in metabolic functions and processes that produce reducing power (NADPH), trehalose and energy, stress rescue and defense systems containing antioxidants (especially, the thiol redox homeostasis systems) and heat shock proteins, and other cellular processes. The heat shock transcription factor 1 (HSF1) of the yeast S. cerevisiae regulates expressions of genes encoding a variety of cell rescue proteins and other functional proteins as well. To better understand the cellular physiological roles of Hsf1, we conducted the hsf1/HSF1 mutant in the thermotolerant yeast S. cerevisiae KNU5377. The hsf1/HSF1 mutant increased the level of hydroperoxide rather than wild type. The hsf1/HSF1 mutant resulted a reduction of the expression of Mn superoxide dismutase (SOD2), catalase (CTT1), glutathione peroxidase genes (GPX1, GPX2, and HYR1), thioredoxin (TRX2), and thioredoxin peroxidase (PRX1 and DOT5), which means HSF1-dependent process. However, the wild-type and cpr1Δ mutant of S. cerevisiae BY4741 as a positive control did not show differences in sensitivity to stress. The down-regulation of G6PDH and cIDH in the cpr1Δ mutant of KNU5377 caused a reduction in the cellular NADPH level. These repressed proteins in the cpr1Δ mutant of KNU5377 were recovered after inhibitor treatment of serine protease B (PMSF) and proteasome (MG132) under MD stress. In addition, the expression of Cpr1p in KNU5377 was regulated by HSF1 under MD stress. We suggest that the CPR1 contributes to the stress tolerance of KNU5377 against various types of environmental stress caused during fermentationthe and has an effect on the expression of antioxidant enzymes and heat shock proteins during MD stress in only KNU5377.
Saccharomyces cerevisiae, potential yeast species used in many fields of industrial fermentation, exhibits significant differences in bread, beer, and wine fermentation resulting from the genomic DNA. To survey the genetic diversity of S. cerevisiae KNU5377 strain, which presents the high fermentation capacity at strain levels, we performed various PCR profiling techniques. Among five S. cerevisiae strains analyzed, genetic differences of KNU5377 were well elucidated by PCR typing of pulse-field gel electrophoresis (PFGE), δ sequences, PCR differentiation using intron splice site primers, random amplified polymorphic DNA (RAPD) analysis with M13 primer, Sau-PCR, and microsatellite analysis, but not by restriction fragment length polymorphism (RFLP) analysis of internal transcribed spacers (ITS). Notably, minisatellite analysis and the nucleotide sequences of the AGA1 gene (YNR044w), the coding a-agglutinin anchorage subunit, proved to be highly polymorphic in KNU5377. The ORF (2519 bp) of AGA1, which contained two blocks of tandem repeat units as serine (Ser) and/or threonine (Thr)-rich block, was longer than that (2178 bp) of S. cerevisiae S288C, the positive control. The current results showed that genetic patterns of KNU5377 were strongly different from the other S. cerevisiae strains. We suggest that KNU5377 might have the different molecular mechanisms involved in the adaptive evolution of yeast traits by the specificities of the molecular basis of the physiological properties such as the generation of evolutionary novelties resulting from significant genetic polymorphism. Menadione (MD), a simplest synthetic quinone used with reactive oxygen species (ROSs) generating agents, causes damages to S. cerevisiae KNU5377. To understand antioxidant defense mechanisms under adaptive stress response, we have performed biochemical analyses and the genome-wide gene expression analysis after exposure to 120 μM of MD with and without adaptation. KNU5377 could survive at higher concentration of MD, 680 μM for 1 h, comparing with S. cerevisiae W303, ATCC24858 as reference strains. Glutathione peroxidase (GPX2), thioredoxin peroxidase 2 (TSA2), and thioredoxin reductase (TRR1) were up-regulated over 10-fold after treatment of 120 μM. Superoxide dismutase (SOD1), catalase (CTA1), glucose-6-phosphate dehydrogenase (ZWF1), glutathione peroxidase (GPX2, HYR1), glutathione reductase (GLR1), thioredoxin peroxidase (AHP1), and thioredoxin 2 (TRX2) were also strongly induced after adaptation. Additionally, transcriptional analysis of those genes was well corresponded with up-regulated protein expression and enzyme activity. Antioxidant defense mechanisms containing SOD1, ZWF and TRX2 maybe orchestrate by the different levels at the same time and function to produce effective defense systems. We also studied the transcriptional regulation using DNA microarray technology to adapt mandione (MD)-induced oxidative stress in a yeast S. cerevisiae KNU5377. Of ~6200 ORFs investigated, approximately 6000 genes were detected. In the MD-treated cells without pre-adaptation, 782 genes showed a > 2 fold higher mRNA level, whereas in the pre-adapted cells up-regulation of 1110 genes identified. Of the up-regulated genes in both cells, approximately 35% belonged to uncharacterized genes (hypothetical ORFs). In particular, the KNU5377 strain represented induction of trehalose -synthesis and/or -degradation genes (TSL1, TPS2, NTH1 and ATH1), genes (FAA1 and CAT2) involved in peroxisome functions, genes encoding reactive oxygen species (ROS)-scavenging systems such as superoxide dismutase (SOD1) and thioredoxin peroxidase (TSA1, AHP1, TSA2, DOT5) and glucose-6-phosphate dehydrogenase (ZWF1) in the both cells. Expression changes of the most genes encoding heat shock proteins (Hsps) such as Hsp104, Hsp90, and Hsp30 were stimulated although the expression intensities were not same. However, gene expression of HSP60 was not corresponded between mRNA level and protein level. As best showed by genome-wide analysis of gene expression, adaptive response to MD stress showed a more strong transcriptional stimulation of the genes involved in metabolic functions and processes that produce reducing power (NADPH), trehalose and energy, stress rescue and defense systems containing antioxidants (especially, the thiol redox homeostasis systems) and heat shock proteins, and other cellular processes.
