The factors that contribute to insulin resistance could increase during aging. In the present study, it was investigated whether ferulate modulated MnSOD and catalase expressions through PI3K/Akt/FOXO signaling during aging. To investigate effects of ferulate on FOXO activity and its regulators with age, aged rats were fed a diet containing ferulate at dose of 3 or 6 mg/Kg/day for 10 days. FOXO activity was decreased in aged rats and stimulated by ferulate. It resulted in the induction of MnSOD and catalase expressions in aging process. PI3K/Akt pathway including p-IRS1, p-PDK-1, p-PTEN, and p-Akt was modulated in ferulate-fed old rats. Induction of PI3K/Akt signaling pathway resulted from age-related increase of insulin levels. In ferulate-fed old rats, the insulin sensitivity in insulin-sensitive organs was improved with the consequence that decreased level of insulin. The amelioration of insulin sensitivity in ferulate-fed old rats was supported by not only decreased glucose levels but also reduced free fatty acid levels. In conclusion, ferulate may have the effect on FOXO transcriptional activity which is response to the expressions of MnSOD and catalase and the inhibition of PI3K/Akt activity through the reduction of insulin levels accompanied with the improvement of insulin sensitivity in insulin-sensitive organs in the aged rats. Significance of the present study is the finding of ferulate for the potential application as an anti-aging agent and also an insulin sensitizer.
