The aggregation characteristics of red blood cells (RBCs) play an important role in the microvascular flow system and increased RBC aggregation has been observed in various pathological diseases, such as thrombosis and myocardial infarction. Although many methods to measure red blood cell aggregation are developed, most of them are not suitable to be used in clinical environment due to various reasons, such as labor-intensive and time-consuming cleaning process. This study stems from the desirability to achieve a suitable measuring technique for measurement of red blood cell aggregation in dynamic condition and to apply for clinical use. A novel measuring technique based on microfluidic slit rheometry using a shear-scanning mechanism and laser backscattering detection is developed, and proposed to assess dynamic RBC aggregation in a disposable microchannel which enables the present system to be easily used in a clinical setting. Two aggregation indices, critical shear stress and critical time, are newly proposed to quantify the dynamic aggregability of red blood cells and experimentally proven by the comparing with some conventional indices. Also, newly-proposed RBC aggregation indices were investigated with varying storage temperature, anticoagulant, temperature, hematocrit and fibrinogen concentration. The present microfluidic application of RBC aggregation offers a quick and easy method in the study of blood rheology and has great potential for point-of-care use.
