This study has evaluated alteration of stress response mechanism related to HPA axis and SAM system using chronic stress-exposured rats. Chronic stress was induced for 15 hours per day (6:00 PM ? 09:00 AM), and this was consecutively repeated for 7 days. After then, animals were allowed to recovery period for 7 days. Experimental groups were divided into 4 groups: control (Ctrl), surgical operation control (OP), restraint stress (Stress) and surgical operation with restraint stress (OP Stress), and each group was subdivided into 5 groups (1, 2, 3, 5 and 7 days) for stress period and 1 group (14 days) for recovery period.
There was a dramatic change of body weight and plasma corticosterone contents in stress-exposured rats. In animals of Ctrl and OP groups, body weight gradually increased during experimental period. In contrast to these, body weight in Stress and OP Stress groups greatly decreased until 7 days after induction of stress, followed by gradual increase. Plasma corticosterone contents in stressreceived animals was significantly higher than that in Ctrl group through whole experimental period and rhythmically changed according to the experimental period. Differential expression of neuronal nitric oxide synthase (nNOS) and c-fos in paraventricular nucleus (PVN) and tyrosine hydroxylase (TH) in locus coeruleus (LC) was examined to clarify the HPA axis response to the repeated restraint stress. nNOS and c-fos in Stress group were significantly increased at 1, 2 and 3 days after receiving restraint stress. At 1, 2 and 3 days, expression of TH in LC of Stress group was significantly higher than that in Ctrl group, and the highest expression was observed at 3 days. This study also showed a significant increase in 5-hydroxytryptamine (5-HT) expression in dorsal raphe (DR) of stressiv received animals. Using Cresyl violet staining method, this study evaluated the changes of pyramidal cell density in hippocampus CA2 area by the repeated restraint stress and observed that cell density in pyramidal cells of stress-received animals was dramatically decreased until 3 days after starting stress induction.
