XTT assay를 통해서 본 실험의 샘플 농도를 100~500 μg/mL로 정하게 되었다. 민들레뿌리추출물의 간 보호 효과를 알아보기 위하여 세포에 300 mM의 에탄올을 처리한 후 열수 추출물(TOH)과 80% 에탄올 추출물(TOE)을 처리하였을 때, 열수 추출물을 처리한 군에서 세포의 생존율이 유의적으로 증가함을 알 수 있었고, 세포내 ROS의 형성 정도를 측정하기 위한 테스트에 있어서도 열수 추출물이 조금 더 높은 효능을 보임을 알게 되어서, 본 실험에 사용할 시료로 TOH을 선택하게 되었다. In vitro 상의 실험을 위해 그룹을 4그룹으로 나누어서 정상군, 에탄올처리군(300 mM), 저농도의 샘플(100 μg/mL)과 에탄올처리군, 고농도의 샘플(500 μg/mL)과 에탄올 처리군으로 나누어서 5일동안 샘플과 에탄올을 처리한 후 지질 과산화물인 MDA 측정한 결과 에탄올 투여군에서 유의적인 증가를 보였으나 시료를 같이 투여함으로써 지질의 과산화 정도가 감소함을 알 수 있었다. GSH함량과 대표적인 항산화 효소인 catalase, GST, GR 및 GPx 측정시에도 에탄올만 투여 하였을 때 보다 시료를 전처리한 군에서 유의적으로 높은 활성을 보임을 알 수 있었다. 이와 같은 결과는 민들레 뿌리 열수 추출물의 처리가 세포의 알코올성 간손상을 억제함을 시사한다. 위 결과를 토대로 in vivo 상의 실험을 위해서, 32마리의 ICR 마우스를 대상으로 같은 방법으로 정상군, 에탄올처리군(5 g/kg bw/day, 저농도의 샘플(0.25 g/kg)과 에탄올처리군, 고농도의 샘플(1 g/kg)과 에탄올처리군, 4그룹으로 나누어서 8일 동안 시료와 에탄올을 처리한 후 마지막 투여 두 시간 후 해부를 통해 혈액과 간, 그리고 신장을 채취하였다. 혈액 분석 결과, 에탄올만 투여한 군에 비해 시료를 전처리한 군에 있어서 간독성 지표인 GOT, GPT, ALPs 그리고 LDH 수치가 감소하였다. 또한, 간과 신장을 상대로 지질 과산화물인 MDA와 GSH함량, 그리고 항산화 효소인 catalase, GST, GR 그리고 GPx를 측정하였을 때에도 에탄올 처리군에 비해서 시료 처리군에서 지질 과산화 정도가 감소하였을 뿐 아니라 항산화 효소들의 활성 또한 유의적인 증가를 보였다. 위의 결과를 종합해 보았을때, 민들레 뿌리의 열수추출물이 알코올로 인한 간 손상에 대한 예방효과가 있음을 확인하였다. 따라서 민들레 뿌리의 열수추출물이 간 손상 예방용 천연 항산화제 개발에 활용될 수 있을 것으로 기대된다.

Nevertheless, the consumption of alcohol carries a risk of adverse health and social consequences related to its intoxicating, toxic and dependence-producing properties. In addition to the chronic diseases that may develop in those who drink large amounts of alcohol over a number of years, alcohol use is also associated with such an increased risk of acute health conditions as alcoholic liver disease (ALD). This study was undertaken to investigate the hepatoprotective effects of the roots of Taraxacum officinale (TO) against alcoholic liver damage by using two experimental models;ⅰ) HepG2/2E1 cells for in vitro, and ⅱ) ICR mice for in vivo. For these tests, TO was extracted by hot water (TOH) and 80% EtOH (TOE). Of two samples, TOE was relatively high in phenolic compounds and flavonoids. To measure the antioxidant activities of TOH and TOE, DPPH radical- and superoxide anion radical-scavenging activities, and TBARS generation inhibitory activities were examined. In DPPH radical- and superoxide anion radical-scavenging activities, TOE presented more high scavenging activities. When their concentrations were 150 μg/mL, DPPH radical scavenging activities of TOH and TOE were 40.7% and 51.0%, respectively. Also, the superoxide anion radical scavenging activities were 4.7% and 17.8%, respectively. However, in lipid peroxidation test, TOH showed the high inhibitory activity. The inhibitory effects on TBARS generation were 37.8% and 28.2%, respectively. To measure the hepatoprotective effects of TOH and TOE in vitro, assays for the ethanol-induced cytotoxicity test by XTT and the formation of intracellular reative oxygen species (ROS) were performed in HepG2/2E1 cells. The pre-treatment for 24 h up to 1,000 μg/mL of TOH or TOE exhibited no cytotoxic effects. To determine the in vitro hepatoprotective effects of TOH and TOE, HepG2/2E1 cells were treated with samples in the presence of 300 mM ethanol(EtOH). Compared to the cells with the treatment of TOE, TOH treated cells showed the high viability, indicating that TOH possessed the preventive effects against the alcoholic damage. Also, TOH inhibited the formation of intracellular ROS. Therefore, TOH was employed to investigate the mechanistic elucidation of its hepatoprotective property. When TOH was treated in HepG2/2E1 cells, the changes in enzymatic antioxidative activities such as catalase, glutathione-S-transferase (GST), glutathione peroxidase (GPx) and glutathione reductase (GR), and non-enzymatic antioxidative activities such as levels of glutathione (GSH) and malndialdehyde (MDA) were examined. The cells were divided into 4 groups: ⅰ) Control (NC), ⅱ) 300 mM EtOH (EC), ⅲ) 300 mM EtOH plus 100 μg/mL of TOH (TOH-L), and ⅳ) 300 mM EtOH plus 500 ug/mL of TOH (TOH-H). The activities of catalase, GST, GPx and GR in EC were significantly lower than those in NC. On the other hand, ethanol plus TOH group stimulated the hepatic enzyme activities and GSH level. Also, the level of MDA which had been increased by EtOH were markedly decreased in the TOH pre-treated group. From these results, we recognized that TOH might be considered as a useful agent in the prevention of alcoholic liver damage induced by oxidative stress. Next, the capability of TOH to protect against ethanol-induced hepatotoxicity and oxidative stress was investigated in the animal model. The ICR mice were divided into 4 groups: ⅰ) group 1 (NC); control mice treated with a saline, ⅱ) group 2 (EC); mice orally received 5 g/kg bw/day of EtOH, ⅲ) group 3 (TOH-L); mice orally received 5 g/kg bw/day of EtOH with 0.25 g/kg bw/day of TOH, and ⅳ) group 4 (TOH-H); mice orally received 5 g/kg bw/day of EtOH with 1 g/kg bw/day of TOH. This test was performed for 8 days. As expected, the administration of EtOH showed the significant hepatotoxicity, which was indicated by drastic increases in the serum GOT and GPT activities. In addition, EtOH treatment produced the high levels of oxidative damages, revealing the significant elevation of hepatic MDA level and the significant reductions of GSH concentration and catalase, GST, GR, and GPx activities, which suggest a role of oxidative stress in EtOH hepatotoxicity. However, pre-treatment with TOH showed a significant protective effect against EtOH-induced hepatotoxicity and oxidative stress in mice. A decrease in the hepatic MDA level confirmed that the treatment with TOH could effectively protect against the hepatic lipid peroxidation induced by EtOH. Also, the antioxidative status was improved by the supplementation of TOH, as evidenced by the elevations of GSH level and antioxidant enzyme activities. Therefore, hot-water extract of Taraxacum officinale (TOH) roots showed the hepatoprotective effects against alcoholic liver damage mediated by alcohol in vitro and in vivo. Based upon these results, it is suggested that the TOH could be used as the useful. hepatoprotectant against alcoholic liver diseases.
