PCR산물에 제한효소 Hpa II 및 Mse I를 이용하여 RFLP분석을 한 결과 차나무 집단간 또는 집단내 차나무 간에 유전적 다양성을 보였다. Hpa II 제한효소를 이용한 RFLP 밴드패턴은 3가지 형태로 구분되었으며, 같은 차나무 집단 내에서도 유전적 다양성이 나타났다. Mse I 제한효소를 이용한 밴드패턴은 6가지의 형태로 다양성을 보였으며, 웅포 집단 차나무의 경우 집단내 다양성이 2가지 형태로 나타나 같은 집단내에서 유전적 변이는 적은 것으로 나타났다. 본 연구에서 사용한 2가지의 제한효소의 결과 차나무의 집단간 또는 같은 집단내의 차나무에서도 유전적 다양성을 확인할 수 있었다. 본 연구결과에 따르면 차나무 분포지역의 식생구조는 집단간 종구성이 다양하고 이질적으로 나타났으며, 많은 지역이 인위적으로 훼손되거나 관리되고 있는 지역임을 확인할 수 있었다. 차나무 엽과 화분의 형태학적 특징들에 대한 유집분석으로 비교한 결과 각 집단간 차이가 있었으며, 엽의 형태적 특징과 화분의 특징이 일치하지는 않았다. The study was conducted to analyze ecological characteristics based on vegetation structure and to classify the variation in leaf morphology and morphological characteristics of pollen among natural grown 22 Tea plant populations in Korea. Also, molecular relationships and genetic diversity were examined to find variations among the populations. Ecological characteristics Species with above 4% of importance value in the study sites were Pinus densiflora, Camellia sinensis, Quercus serrata, Acer palmatum, Torreya nucifera, Pinus rigida, Styrax japonica. Various diversity indexes in the tea plant populations were measured as to the Shannon diversity index (H') ranged from 0.70 to 1.28, Hmax' ranged from 1.15~1.59, Evenness (J') ranged from 0.51 to 0.88, Dominance (λ) ranged from 0.12 to 0.49. Indices of population similarity showed that the composition of species among the populations were very complex and heterogeneous. Total 126 plant species were found in the study sites and there were positive relationships among 43 species that had the impotance value above 5%. By the cluster analysis, community types were classified as communities with Quercus serrata-conifer, deciduous, Quercus serrata-Acer palmatum,-Chamaecyparis obtusa, Quercus - -Acer palmatum,, Pinus densiflora, Torreya nucifera. Leaf morphology Correlations among the leaf morphological characteristics of the tea plant populations ranged from the coefficient value -0.36 to 0.89. The numbers of left and right tooth at the leaf margin had the highest correlation coefficient value (0.89). Whereas the index of leaf shape and leaf width had the lowest value (-0.36). There were positive correlations at 1% level between the lengths from the leaf bottom to the first teeth of both side, and between leaf length and leaf width. As results of the analysis in leaf morphology, the tea plant populations were sorted into 4 groups. The first was Sunyang population, the second was Taean-sa population, the third was Gosa population, and the forth group was categorized to 3 subgroups. Sunam-sa, Songkwang-sa, Bulgab-sa, Unheung-sa, and Bongdae-san were the first subgroup, Yeongok-sa, Hwaum-sa, Baekyang-sa, Borim-sa, Chunkwan-san, Chuneun-sa, Dasan-chodang, Saerong, and Wongpoe were the second subgroup, and Gambul, Gumsan-sa, Siki-dong, Sunun-sa, Bori-am were the third subgroup. The pollen shapes were a isopolar monads and subprolate or rarely prolate spheroidal in equatorial view. The mean length of polar axis were 41.6㎛. The type of arrangement of the pollen grains were tetrahedral and the surface were reticulately patterned. 6 morphological characteristics of pollen grains were analyzed for the correlations, and overall the coefficient values were ranged from -0.20 to 0.55. Six kind of restriction enzyme profiles obtained from digested with restriction endonuclease Mse II and shown two kind of restriction enzyme profiles collected from same region wild tea. The results of RFLP analysis indicated that wild tea shown genetic diversity among the tea plant populations, also among the same region. The results of this study showed that the tea plant populations were very complex and heterogeneous in the composition of plant species. Tea leaf morphology and pollen morphology showed the variations among the populations, while there was no relationship between leaf morphology and pollen morphology. The results of RFLP analysis indicated that wild tea shown genetic diversity among the tea plant populations, also among the same region.
