Extracellular ATP (exATP) has significant functions in the growth of plant. I investigated environmental conditions which induce an increased exATP concentration by Arabidopsis seedlings grown for 15 days. Arabidopsis seedlings had increased extracellular ATP (exATP) concentrations transiently in extracellular matrix (ECM) under hypertonic stress of MgCl₂, Mg(NO₃)₂, and NaCl. The increased transcription levels of two apyrase genes, AtAPY1 and AtAPY2, in accordance with exATP accumulation suggests active regulation of exATP concentration. Arabidopsis seedlings subjected to hypertonic stresses survived after incubation with of β,y-methyleneadenosine-5'-triphosphate, which usually causes cell death through competitive exclusion of ATP. It supports that the enhanced viability is due to accumulated exATP. Another possible function of exATP is as a defense against H₂O₂-evoked cell death like in animal systems. The concentration of hydrogen peroxide was increased by hypertonic stress with a time-dependent pattern similar to exATP. The mRNA levels of exATP inducible genes (AtMAPK3, AtACS6, and AtERF4) and the reactive oxygen species inducible gene (AtPAL1) were also increased by hypotonic stresses. Thus, exATP accumulation seems to be a regulatory mechanism for hypertonic stress response in Arabidopsis seedlings. After ATP, S-adenosyl-L-methionine (SAM) is the major high energy intermediate in the cell. SAM is implicated in a wide range of cellular physiological processes. SAM is synthesized from methionine and ATP in a reaction catalyzed by the enzyme SAM synthase (SAMS). I have used transgenic technology in an attempt to evaluate their potential for mitigating the adverse effects of several abiotic stresses in Arabidopsis. Sense construction of full-length cDNA for S-adenosylmethionine synthase (SAMS) from solanum (S. brevidens) was introduced into Arabidopsis (A. thaliana Col-0) by Agrobacterium tumefaciens-mediated transformation. Several transgenic lines overexpressing SbSAMS gene under the control of cauliflower mosaic virus 35S promoter accumulated high concentration of SAM by 1.7 to 2 times than wild-type plants. The transgenic Arabidopsis also showed significant difference in the development of yellow in the leaves. At a given age, the natural senescence was produced in transgenic lines prior to control plants. SAMS and ACS transcripts were upregulated in a gene-specific manner in response to exogenous application of NaCl and mannitol stresses and treatment with IAA and ethephon. SbSAMS-overexpressing seedlings were resistant to NaCl and mannitol and were response to IAA and ABA. Based on these observations, I concluded that exATP and SAM may help to protect Arabidopsis from stresses through a similar mechanism for wound under the abiotic/biotic stresses
