Since the primary structure of cytochrome P450 BM3 (CYP102A1) from Bacillus megaterium is homologous to that of mammalian P450, it represents an excellent model for studies of the mammalian enzymes. Recently, several mutants of bacterial P450 BM3 have been found to metabolize various drugs through reactions similar to those catalyzed by human P450s. It has therefore been suggested that P450 BM3 may be used to produce large quantities of the metabolites of human P450-catalyzed reactions. During studies on the site-directed mutagenesis based on the 3D structure of BM3, several mutants at the substrate-channel and active site were observed to oxidize several human substrates with higher activities. Several mutants are found to include the amino acid residue of Leu188, which is located around the entrance of substrate-channel. In this study, I examined the role of Leu188 residue by comparing to 19 mutants with different amino acids. A set of 19 Leu188 mutants was made by site-directed mutagenesis and expressed in Escherichia coli to compare enzymatic and biochemical properties to those of the wild-type BM3. Only L188T didn't show an apparent CO peak although it was expressed in E. coli. Other 18 mutants were expressed in E. coli and purified from cytosolic fractions. All of the mutants show apparent catalytic activities towards several typical P450 substrates including fatty acids, 7-ethoxycoumarin, and 12-pNCA. Some mutants show higher catalytic activities than that of wild-type BM3. I have discussed a possible role of Leu188 residue in view of structure and function of P450 BM3.
