1,3-Dichloro-2-propanol (1,3-DCP) is used in high volumes as an intermediate in the production of epichlorohydrin, a monomer used widely in the production of epoxy resin. The major adverse effects of 1,3-DCP in humans are hepatotoxic effects. Potential 1,3-DCP toxicity has been extensively studied over the past several decades in both short-term and long-term animal studies. The possible adverse effects of 1,3-DCP on the reproductive and developmental outcomes have also been examined using experimental animals, but the potential adverse effects of 1,3-DCP on pregnant dams and embryo-fetal development are unclear. The present study investigated the potential adverse effects of 1,3-dichloro-2-propanol on pregnant dams and embryo-fetal development after maternal exposure on gestational days (GD) 6 through 19 in Sprague-Dawley rats. The test chemical was administered to pregnant rats by gavage at dose levels of 0, 10, 30, and 90 mg/kg per day. All dams were subjected to a Caesarean section on GD 20, and their fetuses were examined for any morphological abnormalities. At 90 mg/kg, maternal toxicity manifested as an increase in the incidence of clinical signs, lower body weight gain and food intake, and an increase in the weights of adrenal glands and liver, and increases in AST, ALT, cholesterol, triglyceride, ALP and bilirubin, and decreases in glucose, albumin and total protein. Developmental toxicity included a decrease in the fetal body weight and an increase in the incidence of visceral variation and skeletal variation. At 30 mg/kg, only a minimal maternal toxicity, including lower food intake and increases in the liver weight, cholesterol and triglyceride, and a decrease glucose was observed. In contrast, no adverse maternal or developmental effects were observed at 10 mg/kg. These results show that a 14-day repeated oral dose of 1,3-DCP is minimally embryotoxic, but not teratogenic, at a maternally toxic dose (90 mg/kg/day) and is no embryotoxic at a minimally maternally toxic dose (30 mg/kg/day) in pregnant rats. It is concluded, therefore, that the developmental findings observed in the present study are secondary effects to maternal toxicity. Under these experimental conditions, the no-observed-adverse-effect level of 1,3-DCP is considered to be 10 mg/kg/day for dams and 30 mg/kg/day for embryo-fetal development.
