However, the detailed mechanisms for this effect have been not well determined. The purpose of this present study was, therefore, to address whether the ethanol extract of Lycopus lucidus Turcz (ELT) has positive inotropic effect in perfusated beating rabbit atria. ELT increased atrial dynamics, stroke volume and pulse pressure, in a concentration-dependent manner, but had no effect on changes in atrial cAMP efflux. ELT-induced positive inotropic effect was blocked by diltiazem, an inhibitor of L-type Ca2+ channels. Blockade of the Ca2+ release from the sarcoplasmic reticulum (SR) by combined treatment with ryanodine, a SR Ca2+ release-selective inhibitor, and thapsigargin, a SR Ca2+ ATPase-selective inhibitor, significantly attenuated the ELT-induced increase in atrial dynamics. Staurosporine, an inhibitor of non-selective protein kinases, had no effect on the ELT-induced inotropic effect. In addition, oleanolic acid (OA), ursolic acid (UA) and betulinic acid (BA), a well known triterpenoid constituents of ELT, also increased atrial dynamics and ANP secretion without alteration of the cAMP efflux levels, respectively. An increase in atrial dynamics and cAMP efflux levels induced by OA, UA, and BA were not attenuated by pre-treatment with nifedipine, an L-type Ca2+ channel blocker. Furthermore, ryanodine and thapsigargin blocked the OA- and BA-, respectively, but not UA-induced increases in pulse pressure and stroke volume in beatring atria. These results suggest that ELT induces a positive inotropic effect through activation of L-type Ca2+ channels and SR system in beating rabbit atria. The present study also shows that the triterpenoid constituents have a positive inotropic effect and accentuate atrial ANP secretion.
