In this study, we investigated the changes in spectroscopic characteristics of dissolved organic matters (DOM) under various conditions. First, several DOM characteristics were measured using water samples collected a pilot-scale water treatment plant consisting of coagulated·flocculation(C/F), filtration, ozonation and granular activated carbon(GAC) process. The highest removal of DOM was observed in C/F and filtration processes as demonstrated by the reduction of dissolved organic carbon (DOC). the lowest Specific UV absorbance(SUVA) value and the highest total carbohydrate content were observed in the sample from ozonation process. and ozonation process increased protein-like florescence(PLF). The DOM fractions with the molecular weight greater than 2000 Da were reduced over 90% after C/F process. SUVA values and total carbohydrate content were well correlated with a ratio of protein-like and terrestrial humic-like florescence. Second, we investigated changes in organic matter (OM) characteristics using the influent and the effluent samples from a wastewater treatment plant (WWTP) adopting a biological advanced processes. Refractory organic matter (R-OM) was defined as the organic carbon concentrations of the samples after 28-day incubation for this study. Compared to the influent, BOD, COD_(Cr), DOC and TOC concentrations were reduced for the effluent. The distribution of R-OM of the effluent was much higher than in the influent. Comparison for the spectroscopic characteristics of the influent versus the effluent revealed that the SUVA, the fulvic-like fluorescence (FLF), the humic-like fluorescence (HLF), the MW values were enhanced in the effluent. No significant changes in the OM characteristics were found for the effluent after the 28-day incubation whereas the degree of the changes were great for the influent. This observation suggests that the changes of the wastewater OM characteristics during the treatment processes in the plant amy be similar to those observed for the 28-day incubation. Third, we investigated changes in spectroscopic characteristics and pyrene binding coefficients of terrestrial dissolved organic matters (DOM) during microbial incubation. The incubation studies were conducted for 21 days using a leaf litter DOM and a soil-derived DOM with an inoculum from a river. After the 21-day incubation, DOC were reduced to 61 % and 51% of the original concentrations of the litter DOM and the soil-derived DOM, respectively. Comparison of the spectroscopic characteristics before and after incubation revealed that the SUVA, FLF, HLF values of the two DOM were enhanced by incubation whereas PLF was reduced. This indicates that more aromatic and humic-like compounds were enriched during the biodegradation process while biodegradable and weak carbon structures were depleted. Irrespective of the DOM sources, SUVA values showed a positive relationship with pyrene organic carbon-normalized partition coefficient(K_(oc)) with a correlation coefficient of 0.97. The FLF and HLF also exhibited good correlations with K_(oc) values although different regression equations were obtained from the two different DOM. Our results suggest that the selected spectroscopic characteristics could be good estimation indices for the changes of the binding reactivity of DOM for hydrophobic organic contaminants during biodegradation process. Lastly, we investigate characteristics of humic substances on the changes in dissolved organic matter (DOM) characteristics by biodegradation using three types of the artificial water samples composed of glucose and Suwannee River fulvic acid (SRFA). The changes of the DOM characteristics were minimal for SRFA during the incubation whereas they were significant for glucose. SUVA, dissolved organic carbon (DOC)-normalized fluorescence intensity, and molecular weight(MW) values of glucose increased, suggesting that such labile organic compounds could be exclusively transformed into more humidified materials by biodegradation. For glucose-SRFA mixture, the selected DOM characteristics were greater than those estimated using the assumption that the individual changes of either glucose or SRFA are conservative for the mixture of the two materials. Our results suggest that the presence of humic substances (HS) may lead to the enhancement of the formation of refractory organic materials during biodegradation of labile compounds.
