갈조류(brown algae) 다당류는 alginate와 laminaran으로 구성되어 있으며, 이를 가수분해하면 바이오 에탄올을 생산 할 수 있는 가능성이 높다. 본 연구에서는 열처리와 산처리 및 효소적 가수분해를 이용하여 갈조류를 당화하고 이를 통한 바이오 에탄올 생산을 확인하고자 하였다. 에탄올 발효에는 Saccharomyces cerevisiae KCCM1129를 이용하였으며, 갈조류를 가수분해하기 위한 균주는 기장지역의 해수에서 분리하였다. 분리한 갈조류 가수분해 균주는 Bacterium antarctica로 확인 되었으며, 이를 이용한 효소적 가수분해에서 다시마를 기질로 이용했을 때 최대 1.90 g/L의 환원당이 생성되었다. 가수분해 효율은 효소적 가수분해 보다 산처리의 효과가 더 좋았으며, 3종의 갈조류 중 산을 이용한 전처리에서 톳에서 최대 4.0 g/L의 환원당이 생성되었다. 그러나 바이오 에탄올의 생성은 다시마에서 가장 높게 나타났다. 따라서 다시마를 이용한 에탄올 생산에 미치는 산 및 염의 영향에 대해 추가로 실험하였다. 다시마를 이용한 최적 전처리 조건에서의 발효 결과 2.09 g/L의 에탄올 생산을 확인 할 수 있었다. 산처리 과정에서 생성되는 염의 영향을 확인한 결과 염 농도의 증가에 따라 에탄올 발효가 저해되는 것을 확인 할 수 있다. 갈조류 성분 중 에탄올 발효에 영향을 미치는 당 성분을 확인하기 위하여 주요 당 구성성분인 alginate, laminaran과 mannitol을 이용하여 에탄올 발효 가능성을 확인 하였다. 실험결과 mannitol만이 열처리에서 최대 3.09 g/L까지 에탄올로 전환 가능한 것으로 확인되었으며, laminaran의 경우 0.1 N HCl을 처리하였을 때 0.15 g/L의 소량의 에탄올 생산이 확인 되었다.

Brown algae polysaccharide, consisting of alginate and laminaran, is usable as high bio-ethanol production if hydrolyzed to monomer unit. The objective of this study was to produce bio-ethanol from brown algae by only heat-treatment, acid-treatment or enzymatic saccharification. Bio-ethanol was produced by Saccharomy cescerevisiae KCCM1129 strains. We isolated a new alginate lyase and laminaran lyase producing microorganism from southern sea of Gijang Bacterium antarctica for the hydrolysis of brown algae. The reducing sugar was obtained 1.90 g/L from Laminarin japonica 20 g/L by the enzyme of Bacterium antarctica. But enzymatic saccharification was lower than heat and acid-treatment, Laminaran japonica optimum pre-treatment was used to derive the ethanol production to 2.09 g/L. Bio ethanol production was higher after acid pretreatment than non-acid pretreatment. Only mannitol resulted in the high production of bio-ethanol(maximum 3.09 g/L) from various polysaccharides of brown algae. In case of laminaran, only 0.1 N HCl pretreatment produced ethanol on very low concentration of 0.15 g/L, and cell growth was higher than other pretreatment.
