노로바이러스는 세계적으로 바이러스성 급성 위장염의 주요 원인으로 알려져 있다. 다른 장내 바이러스와는 달리 세포 배양이 불가능하고, 염기 서열의 변화가 심해 분석에 어려움을 겪어 왔다. 본 연구에서는 울산지역 단체 급식소의 지하수를 대상으로 RNA 추출방법과 One step RT-PCR의 최적 조건을 비교하고 이를 바탕으로 32개 샘플의 노로바이러스 오염 실태 조사를 수행하였다. 현장 채수에서 측정한 항목 중 탁도가 높았던 지점에서 노로바이러스 검출이 집중되었고, 계절별로 분석해본 결과 9월(가을)에 탁도가 가장 높았는데 이는 여름철 강수로 인해 수질이 급격히 저하되면서 나타난 결과라고 생각한다. pH는 6.9∼7.6 범위에 90%이상의 샘플이 분포되었고, 다른 계절에 비해 여름에 pH가 높게 나타났다. 필터카트리지의 탈리 단계에서는 1.5% Beef extract(pH 9.5)를 사용하여 회수하였고, 1M HCl을 이용한 유기 응집 농축법을 이용하였다. RNA 추출 방법은 TRIzol과 상용화된 kit를 이용한 방법, 그리고 magnetic bead를 이용하여 비교한 결과, magnetic bead로 추출한 RNA의 PCR 결과가 민감도가 높고 가장 효율적인 결과를 나타냈다. 지하수 중 노로바이러스 분석 결과 2개 지점에서 GI Type의 노로바이러스가 검출되어 6.8%의 검출율을 보였고 우리나라에서 GII Type의 검출율이 GI Type 보다 높게나온 결과와는 상반된 데이터를 얻었다. 노로바이러스가 검출된 단체급식소에서 식중독은 발생하지 않았고 추후 끓인 물 제공과 염소 소독기의 농도 조절, 조리 종사자에 대한 위생 교육 강화 등의 개선 사항이 수행되었다. 본 연구를 통해 지역적 오염 실태를 파악하고 개선하여 식품 위생 안전의 확보를 위한 기초 자료가 될 것으로 사료된다.

Norovirus is known worldwide as an important cause of acute gastroenteritis. Different from other intestinal viruses, the cell culture of norovirus is impossible and its change of base sequence is severe, thus making its analysis very difficult. In this study, the investigator compared optimal conditions between the RNA extraction method and the one step RT-PCR, focusing on the underground water at group dining facilities in Ulsan area. And based on the results of the comparison, the investigator examined the current status of norovirus contamination for 32 samples. Among measured items at on-site sampling of water, the site of which turbidity was higher was also severe in the detection of norovirus. As for seasons, the turbidity in September (autumn) was the highest. It seems to be because of that due to the rainfall in the summer the quality of water deteriorated rapidly. As for pH, more than 90% of the samples were placed in the scope of 6.9~7.6. Comparing to other seasons, the pH in summer was higher. In the desorption stage of filter cartridge, it was collected by using the 1.5% Beef extract (pH 9.5) and the 1M HCl-oriented organic flocculation concentration was used. As for the method of RNA extraction, the investigator used TRIzol and commercialized kit-used method. Comparing it by using the magnetic bead, the result of PCR of RNA, extracted with magnetic bead, was higher in sensitivity and it was most efficient. GⅠ type norovirus was detected in two sites of underground water, thus presenting a 6.8% of detection rate. This datum was contradictory to the domestic result that the detection rate of GⅡ was higher than that of GⅠ.
