Botulinum neurotoxin (BoNT) produced by Clostridium botulinum is known as a most potent biological neurotoxin in nature and causes the paralytic disease called as “botulism” by blocking the release of neurotransmitter. Botulism is rare but is of concern because of its high mortality rate. Nevertheless, the study on cell-immune responses to BoNT has been little reported until now. Therefore, the aim of our study was to characterize the immune responses on botulinum neurotoxin type A (BoNT/A) in mice using BoNT/A toxoid which has been classically used for protection against botulism. Two susceptible mouse strains, BALB/c and C57BL/6, were immunized three times with 5 μg BoNT/A toxoid absorbed with alum and mice inoculated with only alum were used as the control. After the last vaccination, the protection effect in mice was confirmed by BoNT/A challenge. BoNT/A-immunized mice showed complete protection against 10,000LD50 of BoNT/A in both strains. Immune responses on BoNT/A were characterized by investigating antibody responses specific to BoNT/A in mouse sera and analyzing cytokine production and cell proliferation in spleen among three experimental groups including immunization, challenge following immunization, and negative control. From results of serum IgG isotyping, it appeared that IgG1 antibodies were strongly induced in both BALB/c and C57BL/6 mouse strains, indicating that BoNT/A predominantly induces Th2-type response in mice regardless of the strains. This is consistent with the results of cytokine profile produced from BoNT/A-stimulated splenocytes. In both strains, all tested Th2 cytokines including IL-4, IL-5, IL-10 and IL-13 were detected from cell supernatants. However, IgG2a/IgG1 ratio as well as IgG1 titer was definitely high in BALB/c mice compared with C57BL/6 mice, which is involved in high serum neutralization antibody titer and long time-maintenance of complete protection against BoNT/A challenge. Also, in BoNT/A-immunized and challenged groups, especially in BALB/c strains, spleen weight was increased, neutralization ability in blood was enhanced, and BoNT/A-specific T-lymphocytes in spleen were proliferated. In conclusion, our data showed that BoNT/A could induce a biased Th2-type immune responses in BALB/c-C57BL/6 murine model. BALB/c mice could evoke Th1-type response together with Th2-type response, resulting in more stable and more effective protection against BoNT/A than C57BL/6 mice. These data suggest that BALB/c might be an ideal candidate for effective murine model for botulinum research.
