A number of studies have described DPP-Ⅳ resistant GLP-1 analogs with chemical modification or amino acid substitution, in which the latter relied largely upon amino acid substitutions in the N-terminal region of GLP-1 to evade proteolytic degradation by DPP-Ⅳ. Additionally, efforts to prolong the half-life of GLP-1 by attaching macromolecules such as albumin or immunoglobulin heavy chains (IgG-Fc), which are found in abundance in serum, have been sought. In this research, I demonstrated that the N-terminal extension of GLP-1 by a single amino acid, Ala or Gly, successfully prevented degradation by DPP-Ⅳ. Additionally, this study attempted to determine whether the N-terminal extension of GLP-1/IgG-Fc fusion proteins confers resistance to DPP-Ⅳ. Biological activity tests of N-terminal extended GLP-1/IgG-Fc fusion proteins were conducted in INS-1 cells to determine IRS-2 expression, as well as ERK1/2 and CREB activation. GLP-1/IgG-Fc fusion proteins have a prolonged half-life in vivo, as determined via an evaluation of pharmacokinetic profiles generated in Sprague-Dawley rats. These results showed that the addition of either Ala or Gly to the N-terminus of GLP-1 conferred resistance against DPP-Ⅳ and had an average 435.4-times longer half-life in vivo. Furthermore, the A-GLP-1/IgG-Fc fusion protein reduces blood glucose levels in db/db mice. These research findings demonstrate that N-terminal extended GLP-1/IgG-Fc fusion proteins have vast potential for the treatment of type 2 diabetes mellitus; most importantly, the A-GLP-1/IgG-Fc fusion protein may prove useful as a potential anti-diabetic therapeutic agent for the treatment of insulin-resistant type 2 diabetes.
