CLEA법으로 생체촉매 BCA를 고정화하여 CO2의 수화반응평가를 진행한 결과 5번의 재사용에서도 고정화된 생체촉매 BCA는 활성이 줄어들지 않았다. 생체촉매는 이산화탄소를 포집하여 재사용할 수 있는 CaCO3을 안정적으로 생성할 수 있어 환경 친화적인 방법이다.

Biocatalysts were immobilized on mesoporous silica through various approaches, including covalent attachment, adsorption, and cross-linked enzyme aggregation (CLEA). The catalytic activities for hydration of CO2 were calculated for free and immobilized enzyme with and without buffer. The effects of pH, temperature, storage stability, and reusability on the biocatalytic performance of CA were characterized by examining para-nitrophenyl acetate (p-NPA) hydrolysis. Lineweaver-Burk relationship was employed to estimate the Michaelis-Menten kinetic parameters for the free and immobilized enzyme. The kcat/ Km values of free and immobilized CA for p-NPA hydrolysis were 873.76 and 680.11 M-1s-1, respectively, The immobilized CA was stable and did not lose any activity after 5cycles. In addition, ever after 30 days, whereas the free BCA showed a decrease in the catalytic activity of 40% of its original activity, BCA-CLEA catalytic activity was retained about 98%. The optimum temperature for the immobilized BCA was found to be 30℃. The immobilized enzyme provided a green, stable, reusable and convenient biocatalyst for CO2 sequestration and produced pure calcite phase CaCO3.
