Recently, various novel biosensors have been fabricated and developed for early diagnosis of various diseases. Fluorescence Resonance Energy Transfer (FRET) is a physical phenomenon between two fluorophores which transfers emission energy from donor to acceptor for excitation. In this paper, FRET phenomena between two Alexa Fluors, quantum dot and dopamine have been investigated. The FRET efficiency dependence on dopamine concentration has been studied for the application of dopamine sensor. Alexa Fluor 594 (AF594) as a donor has been labeled to protein A (PA) which was conjugated with anti-dopamine labeled with Alexa Fluor 546 (AF546), used as an acceptor. FRET phenomenon was confirmed by the fluorescent emission spectra change by varying the concentration ratio of the fluorophores. The FRET efficiency strongly depends on dopamine concentration. Carboxyl quantum dots have been immobilized on the surface of polystyrene well plate by the EDC/sulfo-NHS coupling reaction, and the fluorescence intensity dependence on dopamine concentration has been studied. The maximum emission peak was observed at 320 nm and 530 nm from dopamine and quantum dots, respectively. As the dopamine concentration increases, emission intensity from dopamine increases, while that from quantum dots decreases. The fluorescent intensity of QDs 530 was quenched as dopamine concentration increased, and this shows that concentration of dopamine can be confirm by QDs 530. The fluorescence intensity change by FRET between QDs and dopamine due to dopamine and QDs reaction can be used to detect dopamine. The optical fiber was tappered by etching with HF solution, and the surface was functionalized to immobilize QDs. The AFM images and PL spectrum confirmed that QDs were immobilized on the surface of the optical fiber.
