The aim of surgical treatment for cardiovascular diseases and peripheral vascular diseases, is to restore blood flow by replacement or bypass with autologous or artificial grafts. However, sometimes it is difficult to harvest sufficient length of autologous grafts or to obtain grafts without pathological change. In such cases, artificial grafts as ePTFE(expanded polytetrafluoroethylene) and Dacron (polyethylene terephthalate fiber) are used to replace or bypass diseased arteries. However, the artificial grafts used at present are nondegradable, even with the occurrence of material related failures and complications, such as stenosis, thromboembolization, calcium deposition, and infection, revealed by long term follow up studies. To overcome these problems, research has been carried out on biodegradable materials using a tissue engineering approach, in which the material is processed as a scaffold to facilitate the adhesion, growth and extracellular matrix production of vascular smooth muscle cells and endothelial cells to generate function.
In this work, we fabricated an artificial blood vessel scaffold(ABV) that was electrospun with an inner polycaprolactone(PCL)-gelatin layer and outer poly(lactic-co-glycolic acid)(PLGA)-chitosan layer. The structure, mechanical properties, biodegradability, cytocom- patibilities, cell attachment, and cell proliferation on each of the ABV layers were investigated by scanning electron microscopy(SEM), tensile testing, incubation in phosphate buffer solution, and endothelial cells(CPAE) and fibroblasts(L-929) culturing. SEM micrograph showed the PLGA-CS cross-linked fibrous mat which contained fibers that ranged from 500nm-1.2μm diameters, was fabricated by mixing the fibers of PLGA and chitosan. The PCL-gelatin cross-linked fibrous mat which also contained same sized fibers, was fabricated by mixing the fiber of PCL and gelatin. The structure of layers was similar to that of extracellular matrix. The electrospun PCL-gelatin/PLGA-chitosan cross-linked mat exhibited the highest tensile strength, stress strain, and lowest tensile modulus, and slowly decreased after incubation for more than 2 months, as assessed by the weight loss method. The PLGA -chitosan and PCL-gelatin were non-toxic and displayed a high biocompatibility, viability, cellular attachment, proliferation for each of L-929 and CPAE through MTT assay and SEM findings.
Using electrospinning 2-ejection, ABV with high surface to volume ratio, and high porosity were fabricated and thought to mimic natural extracellular matrix and thus promote cell adhesion, migration and proliferation. And ABV assembled through electrospinning had so high tensile strength that was enough to be used for artificial blood vessel.
