Senescent cells express several senescence phenotypes such as an increase of reactive oxygen species, SA β-gal activity, and mitochondria and lysosome contents as well as a decrease of mitochondrial membrane potential. Among these senescent traits, the origin of SA β-gal activity is known to be the lysosomal β-galactosidase activity which is elevated in senescent cells. An increase of SA β-gal activity was also reported in cells either serum-starved or cultivated at a high confluency. However, how are the content and activity of lysosome regulated in these cells is totally unknown. In current study, we observed that the lysosome content and β-gal activity are increased in the serum-starved cells and examined the underlying reason for these changes. It appeared that, at early time points, the lysosome content increases possibly by increased lysosome biogenesis as judged by an increase in the mRNA levels of the various lysosomal proteins and in the precursor forms of Cathepsin B, a lysosome constituent protein. At later stage, however, the increase is likely attributed in large part to the accumulation of autophagolysosomes which have low hydrolysis capability as judged by the appearance of large structures in which LC-3 and lysosome signals colocalized. A possible contribution of the lipofuscin-containing residual body seems to be minimal as judged by the lack of the effect of N-acetyl cysteine, a potent antioxidant which caused a strong attenuation of the lipofuscin-orignated autofluorescence. Overall, our results indicate that cellular lysosome content increase due in part to an increase in the lysosome biogenesis and to an accumulation of autophagolysosome in cellls under serum starvation and suggest the possibility that lysosome biogenesis may happen in certain cellular conditions.
