Recombinant proteins of two GMO markers, neomycin phosphotransferase II (NPTII) and cucumber mosaic virus coat protein (CMV CP), were produced in E. coli. After purification by affinity chromatography, their structural conformation and binding affinities to the respective reference antibodies were characterized. The results revealed that the recombinant proteins produced in this experiment were identical with commercially available reference proteins. These recombinant proteins were used to raise polyclonal and monoclonal antibodies for detecting GMOs harboring either markers. For this purpose, the purified antibodies were applied to sandwich ELISA system to detect and quantify GMO marker proteins, identifying that these antibodies readily discriminate between GMOs and non-GMOs. Moreover, detection capacity of marker proteins were defined to 1%, exceeding over the upper limit of standard in Korean provisions. Those well-characterized marker proteins and antibodies can be used as valuable immuno-reagents in developing analytical systems, such as biosensors and biochips, to measure quantities of GMOs.
