In part I, a rapid multiresidue screening and generic confirmation method were developed using ultra-fast liquid chromatography/tandem mass spectrometry (UFLC-MS/MS) for high throughput efficiency in doping analysis. The urine samples were extracted by two consecutive liquid-liquid extractions at the different pH after enzymatic hydrolysis and analyzed by UFLC-MS/MS with the fast polarity switching and the time-dependent selected reaction monitoring (EZ-method). Taking advantage of higher peak resolution and very narrow peak width of UFLC, total 221 substances from diverse classes on the prohibited list could be successfully identified within a short analytical time of 10 min. Doping analysis can be achieved with single UFLC-MS/MS method for all prohibited substances except for some of anabolic steroids detected only by the gas chromatography-mass spectrometry (GC-MS) method due to hard ionization in electrospray ionization (ESI) source. In part II, a novel gas chromatography-electrospray ionization/orbitrap mass spectrometry (GC-ESI/HRMS) was developed for anabolic steroids. GC and ESI source were connected by self-made heated column transfer line. The interface is simple and can easily be set up in any mass spectrometer equipped with an ESI source, which is the most commonly used ion source in LC-MS instrument. The GC-ESI/HRMS system was optimized to increase ionization efficiency and was applied for 14 anabolic steroids. Methanol contained 0.1% formic acid was used as the ESI solvent. The protonated molecular ion ([M+H]+) was observed in the GC-ESI/HRMS spectra of steroids containing a ketone group or nitrogen. In the spectra of steroids containing only a hydroxyl group, [M+H-H2O]+ and [M+H-2H2O]+ ions without [M+H]+ ion were observed. In GC-ESI/HRMS, two ionization mechanisms are possible. First, the gas-phase analyte eluted by GC is dissolved into the charged droplet generated by ESI, and then the ionization process occurs by ion evaporation. Second, a proton is transferred from protonated solvent to gas-phase analyte. In this study, there are many consistencies in the spectrum between GC-ESI/HRMS and LC-ESI/MS, which strongly suggests that GC-ESI/HRMS mainly follows ion evaporation mechanism. A novel GC-ESI/HRMS provides the high chromatographic resolution and the soft ionization process. The technique could be applied to identify anabolic steroids in the future.
