Background and purpose: Lactate has been considered as a fatigue factor, inducing acidosis during exercise. The balance between oxidative stress and antioxidant is associated with controlling cell damage and prevention during exercise. Previous studies suggested that plasma lactate has an antioxidant effect on oxidative stress, such as superoxide and hydroxyl radical. However, the effect of lactate on the oxidative stress, which plays an important role in oxidation-reduction state during exercise, has not been studied yet.
The purpose of this study was to examine the relationship among the level of blood lactate, oxidative stress and antioxidant during high-intensity exercise.
Methods: Thirty healthy men majoring in physical education were recruited to participate in this study. Subjects performed exercise graded testing to get their maximum capacity of oxygen consumption with Bruce protocol. The intensity was set to be 80% of VO2max based on the graded exercise testing, then the co-relationship among the level of lactate, oxidative stress (hydrogen peroxide; H2O2)and antioxidant (trolox) were examined at resting, exercise 5 min, exercise 10 min and recovery state. The results were analyzed by repeated ANOVA, paired t-test, and pearson co-relation methods using SPSS 12.0 program.
Results: The level of lactate was significantly increased during exercise and showed the highest level at 10 min. Contrast to lactate, the levels of H2O2 and trolox were not significantly increased by 80% VO2max exercise. In the highest lactate concentration, H2O2 concentration was decreased, and trolox level was not changed during exercise.
In the relationship using pearson's method, all factors do not have co-relationship in the period matched blood sampling.
Conclusion: According to the result of this study, it is concluded that lactate can scavenge the concentration of H2O2 during high intensity exercise. Further studies are needed to identify specific mechanisms that show how they work.
Keywords: Lactate, Oxidative stress, Antioxidants, Hydrogen peroxide (H2O2), Trolox.
