Neurohormesis refers to process in response to a moderate level of stress by enhancing their ability to resist a more severe stress that might be lethal or cause dysfunction or diseases. Neurohormetic phytochemicals including resveratrol, sulforaphane, curcumin, and catechins protect neurons against injury and disease. Naphthaquinone such as juglone and plumbagin showed a robust hormetic stress response. However, the possibility that subtoxic dose of naphthazarin (5,8-dihydroxy-1,4-naphthoquinone) may protect brain diseases via activation of adaptive stress response pathway in the brain has not been investigated. In this study was examined neurohormetic effect of subtoxic dose of naphthazarin in Parkinson’s disease model. Subtoxic dose of naphthazarin enhanced movement ability, prevented loss of dopaminergic neuron, and attenuated neuroinflammation in MPTP-induced Parkinson’s disease model. It was found that neuroprotective effect of naphthazarin is mediated by suppression of astroglial activation in response to MPP+ treatment. In conclusion, in view of its hormetic effect on neuroprotection it is suggested that naphtazarin could be beneficial in treatment of aspects of Parkinson’s disease and other neurodegenerative diseases associated with neuroinflammation.
