Realtime-PCR and western blot analysis together with immunofluorescence assay all showed that the expression level of annexin A5 increased significantly by the short term incubation with cisplatin at relatively low level in both human and rat kidney epithelial cells. In regard to the mechanism of cisplatin-induced kidney cell death, AIF release into the cytosol was observed and the underlying mechanism was identified as VDAC oligomerization. VDAC is shown to partly mediate cisplatin-induced apoptosis via the formation of VDAC oligomer pore at mitochondrial outer membrane through which apoptogenic proteins are released into the cytosol. To understand the functional significance of annexin A5 up-regulation in kidney cell death with regard to cisplatin, we used small interfering RNA-mediated approach to knockdown annexin A5 in human kidney epithelial cell line. Annexin A5 depletion led to increased cell viability compared to the control group and significantly, reduced VDAC level. In conclusion, our data suggest that annexin A5 up-regulation plays a crucial role in cisplatin-induced nephrotoxicity by mediating mitochondrial apoptotic pathway via regulation of VDAC.
