본 논문은 마이크로 제작공정과 PDMS (polydimethylsiloxane) 캐스팅을 이용한 마이크로 플루이딕 디바이스의 제작 및 마이크로 플루이딕 디바이스의 가변 초점 유체렌즈와 마이크로 입자 제조로의 응용에 대한 연구를 기술한다. 본 논문은 첫째, 마이크로 유체기술을 이용한 가변 초점 유체렌즈의 제조 및 광학 스캐너로의 응용과 둘째, 플로우 포커싱 마이크로 플루이딕 디바이스 (flow focusing microfluidic device) 의 제조 및 하이드로겔 마이크로 입자 제조로의 응용, 셋째, 마이크로 캐스팅용 하이드로젤몰드 제작으로 구성된다. 첫째, 가변 초점 유체렌즈 부분에서는 가변 초점 유체렌즈의 설계, 포토리소그라피를 이용한 폴리머마스터 몰드의 제작 및 PDMS 캐스팅을 통한 유체렌즈를 제작하였다. 캐스팅과정에서 핫프레스(hot press)공정을 사용하여 마이크로 채널과 멤브레인을 일체형(single body)으로 제작하였으며, 또한 유체렌즈 멤브레인의 두께를 균일하게 유지하였다. 제작된 렌즈의 마이크로 채널을 통하여 주입된 유체의 압력에 의하여 멤브레인이 변형하여 유체렌즈의 형상이 변형되며, 이에 따라 렌즈의 초점거리가 바뀌게 된다. 주입된 유체압력에 따른 렌즈형상 변형을 측정하였으며, 초점거리 및 조리개 수치를 이론적으로 계산하고 광학테스트를 수행하여 비교하였다. 제작된 유체렌즈의 응용으로써, 가변 초점 유체렌즈를 이용한 레이저 스캐닝에 관한 연구를 수행하였다. 유체렌즈를 헤미실린더(hemicylinder)형으로 설계, 제작하고 이를 이용한 레이저 빔을 시트(sheet)형으로 바뀌는 빔 형상 변화 특성을 조사하였다. 또한 두 개의 헤미실린더 유체렌즈를 직교방향으로 정렬하여 레이저 빔으로부터 시트레이저 형성 및 이의 스캐닝 특성을 살펴보았다. 둘째, 플로우 포커싱 마이크로 플루이딕 디바이스에서는 유체장치의 설계, 포토리소그라피와 PDMS 캐스팅을 통한 장치 제작공정과 이를 이용한 마이크로 입자 제조 특성에 대하여 기술하였다.마이크로 채널 내에 두가지 서로 다른 유체상(phase)을 동시에 흘려주면서 발생하는 플로우 포커싱 현상을 이용하여 PEGDA(poly(ethylene glycol diacrylate))와 PEGMEA (Poly(ethylene methyl ether acrylate))의 마이크로 드롭렛(droplet)을 형성하고 이를 광중합하여 마이크로 입자를 제조하였다. 마이크로 채널에 공급되는 유속, 채널 형상, 하이드로젤 농도 등 제조과정의 주요인자에 따른 드롭렛 형성특성을 조사하였다. 또한 기능성 입자가 담지된 마이크로 입자를 제조하였다. 셋째, PDMS 캐스팅을 위한 마스터 몰드 제작을 위하여 하이드로젤을 이용한 몰드제작에 관한 연구를 수행하였다. 하이드로젤 디스펜싱, UV 조사, 그리고 린싱 공정을 통하여 마이크로 구조의 제작이 가능하였으며, 본 방법은 타 공정에 비하여 공정이 간단하고 빠른 장점을 가진다. 제작된 하이드로젤 구조는 PDMS 마이크로 플루이딕 디바이스 제작을 위한 몰드로 사용될 수 있으며, 두께 1mm이상의 마이크로몰드 제작이 가능하였다.

Development of the new strategies for the fabrication and application of the microfluidic devices has been described. In particular, this thesis focused on: i) the fabrication based on microcasting and application of the microfluidic devices as the micro optofluidic devices, ii) the fabrication based on microcasting and application of the microfluidic devices to prepare hydrogel microparticles and iii) the development of a novel method to prepare a master mold for microcasting. First, the new strategy for the fabrication of the microfluidic lens using poly(dimethylsiloxane) heating pressuring polymerization has been developed. Using this method, a microfluidic lens body can be prepared by a single, simple and fast process. The fabricated lenses were investigated to determine the characteristics such as lens curvature change, focal length, numerical aperture and the change of the laser beam after illuminating through the lens under various applied pressures. The relative functions of the focal length and numerical of the hemispherical microfluidic lens with applied pressure were investigated. The characteristics of the hemicylindrical microfluidic lens have been studied. The light scanning mechanism of the orthogonal system of the two hemicylindrical microfluidic lens, which can be really meaningful has been investigated. Second, fabrication of the hydrogel microparticles using the devices which based on the microfluidic system has been developed. The micro flow focusing devices were prepared to do this purpose. Using these devices, poly(ethylene-glycol) microparticles were fabricated. The relationships of the experimental conditions and microparticle formation as the size and shape of them were determined. The strategies for the preparation of the functional elements laden hydrogel microparticles, which can be applied in medical and chemical analysis technical fields have been investigated. Third, a novel method to prepare a very thick master mold for poly(dimethylsiloxane) (PDMS) microcasting was investigated by using hydrogel ultra violet (UV) curing process through a film mask. A simple process of dispensing of hydrogel, UV curing through a photo mask, and rinsing enable construction of micro-hydrogel structures in a fast manner. These hydrogel structures can be used as a mold for PDMS microcasting for PDMS fluidic chip fabrication. This method allows fast construction of very thick microstructures more than 1 mm. The characterizations about vertical side wall and adhesion enhancement between substrate and microstructures were studied. The application of PDMS fluidic chip, which was prepared from the hydrogel mold by PDMS casting, into some fluidic flow rate tests were demonstrated.
