Electrospun scaffolds have been widely used in tissue engineering due to their similar structure to native extracellular matrices (ECM). Their unique fibrous structures promote attachment and proliferation of cells on the scaffolds. However, one of the obstacles limiting the application of electrospun scaffolds for tissue engineering is the nano-sized pores, which inhibit cell infiltration into the scaffolds. To overcome this limitation, we developed small diameter vascular grafts in vitro and in vivo by seeding and culturing fibroblasts on the electrospun scaffolds and constructing a 3D vascular type graft. We fabricated a small diameter vascular graft using Cell Matrix Engineering with fibroblasts and electrospun PLCL scaffolds. Their diameters were 4.1mm (±0.1mm) and they maintained 3-D tubular structures. Also, we compared the mechanical and biological properties of various vascular grafts with electrospun PLCL sheets of different thicknesses. In these experiments, the vascular graft made with thin sheets were shown to result in better cell proliferation and attachment than grafts made with thick sheets because the thin layer allowed for more efficient mass transfer and better permeability than the thick layer. Vascular grafts from thin sheets by Cell Matrix Engineering offered favorable environments to fibroblasts. Culturing under physiological pulsatile flow conditions was demonstrated in this work. These dynamic conditions provided improved mass transport and aerobic cell metabolism. Therefore, the Cell Matrix Engineered vascular graft described in this work holds great promise for use in overcoming the limitations associated of conventional scaffolds for clinical applications.
