PPARs의 발현은 대조군, LPS (5 mg/kg) 처리군, fenofibrate 전처리하고 LPS 처리군 및 fenofibrate (100 mg/kg) 구강 투여군의 마우스 성체의 뇌 조직에서 RT-PCR, 웨스턴 블렛, 면역조직화학염색법을 이용하였고 혈장의 친염증성 사이토카인의 발현은 면역효소분석법 (ELISA)으로 측정하였다. Fenofibrate에 의해서 LPS에 의해 증가된 친염증성 사이토카인인 IL-1b, IL-6 및 TNF-a의 발현은 혈장과 뇌 조직에서 현저히 억제되었고 한편, LPS에 의한 염증반응의 결과 지방산 산화와 같은 지질대사 및 관련된 유전자의 발현이 변경되는 것으로 알려졌다. 따라서 연구에서는 LPS에 의한 염증성반응에 의해 마우스 뇌 조직에서 PPARs (PPARα, PPARβ/δ 및 PPARγ)와 PGC-1이 mRNA와 단백질 수준에서 현저히 감소하였고 fenofibrate에 의해서 회복되는 것으로 나타났다. 또한 PPARs의 증가된 활성화에 의해 LPS 처리군의 뇌조직에서 페록시좀의 수와 효소적 활성과 관련된 ABCD3 (PMP70), ACOX1 및 Catalase의 발현 감소가 억제되었고 뇌조직에서 항산화 능력의 증가되어 LPS 투여에 의한 지질산화와 관련된 3-NT와 4-HNE의 발현은 뇌조직의 조직표본에서 현저히 억제되었다. 따라서, fenofibrate의 항염증 및 항산화적 특성은 파킨스병과 알츠하이머병과 같은 진행성 신경질환을 억제하는데 유용할 것이다.

The objective of the current study was to investigate whether the increased expression of PPARs with fenofibrate treatment was altered in the immune system of brain tissues after lipopolysaccharide (LPS) intraperitoneally injection in BalB/c mice. Present study also investigated the protective effects of fenofibrate, PPAR-α gonist, against the reduced expression of transcriptional factors (PPARs and PGC-1) and these specific target genes, including ABCD3 (PMP70), ACOX1, and Catalase, and the increased production of lipid peroxidation such as 3-nitrotyrosine (3-NT) and 4-hydroxynonenal (4-HNE) after LPS administration. PPARs expression were investigated in the brain tissues of adult mice after controls (sterile saline, n = 6). LPS injection (5 mg/kg BW, n = 6) with pretreated fenofibrate or alone, and fenofibrate (100 mg/kg BW, n=6), by using reverse-polymerase chain reaction, Western blot analysis, and immunohistochemistry. Plasma pro-inflammatory cytokines were also assessed by ELISA assay. Fenofibrate have protective effects in increase of LPS-induced pro-inflammatory cytokines, inclusing IL-1β, IL-6 and TNF-α in serum and brain tissue, repectively ELISA assay and RT-PCR and Western blot analysis. However, LPS-induced inflammatory response was well known to induce alterations in lipid metabolism such as fatty acid oxidation (FAO) following the changes of gene expression. Then, LPS-induced proinflammatory response decrease the mRNA and protein level of PPARα, PPARβ/δ, PPARγ, and PPARγ coactivator 1 (PGC-1) in mouse brain tissue. However, fenofibrate pretreatment markedly suppresses the reduction of PPARs and PGC-1 mRNA and protein level. Moreover, the increased activation of PPARs abrogated the reduction of ABCD3 (PMP70), ACOX1, and Catalase expression and ,then, the increase of antioxidative capacity in brain tissue blocks the production of lipid peroxidation such 3-NT and 4-HNE after LPS administration. Therefore, the anti-inflammatory and anti-oxidative properties of fenofibrate may be useful for ameliorating the progression of neuronal diseases, such as Parkin's and Alzheimer's diseases.
