MIA 접종으로 유발 시킨 골 관절염 쥐의 혈액 내, 면역체계의 대표적인 염증 반응 물질인 TNF-α에 대한 관찰에서 중강도 운동, 고강도 운동에서 모두 변화된 양상을 보였다. 하지만 저강도 운동에서는 변화를 보이지 않았다. 그리고 골 관절염의 혈액 속 단백질에 대한 관찰에서는 질량분석기를 사용 하여 분석 하였다. 분석된 결과에서 총 27개의 단백질을 볼 수 있었고, 그 중 17개의 증가된 단백질과 10개의 감소된 단백질을 관찰 할 수 있었다. 17개의 증가된 단백질 중 9개는 운동 후 감소된 양상을 보였고 10개의 감소된 단백질 중 5개는 운동 후 증가되는 양상을 보였다. 증가된 단백질 중 C9 단백질을 사용하여 결과를 확인하였다. C9 단백질은 MIA 주입 후 모든 군에서 증가하였고, 운동 후 중강도 운동군에서 유의하게 감소되는 경향을 보여 주었다. 저강도와 고강도 운동군에서는 MIA 주입 후 유의한 감소를 보여주지 않았다. 결과적으로 운동이 골 관절염을 유발 시킨 쥐의 혈액 속 단백질들을 변화 시킬 수 있다고 생각되고 적절한 강도의 운동을 적용시킴으로써 골 관절염의 예후에도 기여할 수 있다고 생각된다.

In the present study, changes of TNF-α expression level related exercise/exercise intensity and serum proteins level (related exercise/exercise intensity) were investigated in rat with OA using western blot. The results showed TNF-α expression level was increased after MIA injection and decreased in middle and high intensity exercise groups than MIA injection group. But low intensity exercise group was no changed in TNF-α expression level compared with MIA injection group. In ESI-LC-MS/MS analysis, total 27 proteins were identified. There were 17 up-regulated proteins and 10 down-regulated proteins. In 17 up-regulated proteins, 9 proteins were increased in MIA injection group than cont. group. On the other hand, 5 proteins in 10 down-regulated proteins were decreased MIA injection group than cont. group. Almost these proteins were correlated acute inflammation and inflammatory proteins. Among the list of up-regulated proteins, C9 protein was used for validation. After MIA injection, C9 protein was increased in 4 groups than cont. group. But only middle intensity exercise group was significantly decreased after Treadmill exercise. In conclusion, TNF-α, C9 protein were changed by exercise/exercise intensity, therefore theses results showed exercise/exercise intensity affected TNF-α expression level and C9 protein level in rats serum with OA. Therefore, treadmill exercise with appropriate intensity can be recommended for OA patient treatment.
