Historical patterns of dispersal and geographic isolation have been considered key components in shaping genetic diversity across landscapes. Two additional processes, including topographical landscape structures and adaptive divergence may also influence gene flow among populations within drainages. The preservation of biodiversity and management strategies for freshwater system should require the information about the components of population dynamics. However, the estimation of the components based on field approximation can be complicated, especially in a structured-deme context, where local populations exist as distinct unit, but are interconnected. Metapopulation genetics and island biogeography theory can be applied to reconstruct contemporary episode of dispersal and microevolutionary processes. Here I use neutral genetic loci to determine the amount of genetic variance kept in local populations and to predict the demographic and evolutionary processes in a metapopulation with Korean chub, Zacco koreanus, in Korea. To accomplish this goal, I carried out three separate projects and allocated the results into three chapters, respectively; (1) genetic confirmation of the existence of three species of Korean chub and their reproductive isolation; (2) genetic structure and within-drainage metapopulation dynamics of Zacco koreanus (NS vs NE); (3) non-natural landscape structures and the influences on the contemporary dispersal. 1. Genetic confirmation of the existence of three species of Korean chub and their reproductive isolation. Korean chub (Zacco koreanus), a freshwater cyprinid native to Korea, is ubiquitous and represents a dominant component of upstream communities throughout much of their range. Considerable uncertainty remains about the taxonomic status, as the possibility that Korean chub represents more than a single species was supported by distinct morphological types, i.e. HK, NS and NE, observed among populations. Although the best identification between these types is still morphology (references), the available method by morphological characters alone is rather cumbersome, necessitating much expertise and practice. I examined phylogenetic relationships among those morphological types and dark chub (Z. temmincki), the most closely related species to Korean chub, with: (i) sequence data from two mitochondrial genes; (ii) microsatellite genotyping. The analysis of mtDNA sequences revealed that three types of Korean chub and dark chub are genetically distinct from each other and consistently compose independent monophyletic groups. The microsatellite data further supports the genetic differentiation among those three types and dark chub and provide little evidence for the introgression between types. These results resolve the taxonomic confusion among the three types and verify that Korean chub and dark chub are distinct species. 2. Genetic structure and within-drainage metapopulation dynamics of Zacco koreanus. The results of microsatellite loci indicated marked genetic divergence between type NS and NE. Both types NE and NS were found in southern-flowing rivers, while only type NE is found in short streams along the southern part of east coast, which flows east into the East Sea. In this chapter, Nakdong drainage basin (southern-flowing rivers) was the focus of my research to find (1) empirical evidence of reproductive isolation between two types and (2) metapopulation dynamics of two types. In several locations where two types coexisted, genetic markers from microsatellite loci were used and little evidence of introgression between two types was found, suggesting that those two types are discrete species. Interestingly, FST values among subpopulations were relatively high in NE rather than NS, indicating that resource partitioning between two might affect the gene flow of NE and/or might be probably due to high genetic drift in NE populations. My findings provide little evidence for the equilibrium-island and isolation by distance models. Instead, non-equilibrium processes including genetic drift associated with population reduction, asymmetric gene flow and recolonization, influence the apportioning of genetic diversity among populations. 3. Non-natural landscape structures and the influences on the contemporary dispersal. The effective size of metapopulation is dependent on the level of population genetic subdivision. My results in the previous chapters suggest that local extinction is likely in the freshwater systems examined unless sufficient numbers of sustainable demes are maintained, with implications for conservation given ongoing species decline and habitat fragmentation. The genetic principles from population dynamics gained can be applied to contemporary evolutionary processes given by non-natural landscape structures in the freshwater systems, i.e. dam. I focused on Woongchon Stream, independent western-flowing drainage, where a gigantic dam exists dividing the populations into up- and down-stream. Although conspicuous genetic divergence was not found between up- and down-stream in genotype frequencies, the analysis based on Bayesian probability suggested dominant contemporary migration was found only from downstream regions as a source, whereas upstream sites only received immigrants or contributed emigrants as a sink. In conclusion, non-natural barriers may significantly affect gene flow pattern and source-sink dynamics, including contemporary genetic subdivision.
