방향족 화합물은 인간 활동의 부산물로써 환경을 오염시키는 물질 중 하나이며, 그 중에서도 페놀은 다양한 산업 활동으로부터 나온 폐수를 통해 방출되는 환경오염물질이다. 일반적으로 아미노산을 생산을 위해 사용되는 대표적 산업 균주로 알려진 C. glutamicum은 방향족 화합물을 분해하여 유용한 아미노산으로 전환시킬 수 있다. 본 연구에서는 C. glutamicum의 오염 물질 생분해 능력에 미네랄들이 어떠한 영향을 미치는지 알아보았다. 이는 다음의 방법을 이용하였다. 첫째, 미생물의 성장과 페놀 분해능에 미네랄이 어떠한 영향을 미치는지 확인하였고, 둘째, 오염된 토양 환경에서 미네랄을 처리하였을 때, 페놀의 분해 효율과 아미노산 생산량의 변화를 분석하였다. 마지막으로 실제 오염된 토양에 적용해 보고자 매몰지 토양에 C. glutamicum을 적용하였다. 이로써 본 연구는 C. glutamicum의 환경 오염 물질의 활용 결과로써 오염된 토양 환경을 복원시키는 친환경 기술로써 이용 가능함을 보여주었다. 본 연구에서는 미네랄이 C. glutamicum의 페놀 활용에 영향을 미치는지 알아보기 위하여 페놀 분해능과 아미노산 생산량 그리고 식물을 이용한 토양 내 독성을 분석하였다. 우선적으로 미네랄 처리에 따른 C. glutamicum의 변화 경향을 살펴보기 위하여 수환경에 적용해보았으며, 그 결과 미네랄은 미생물의 성장과 페놀 분해능에는 아무 영향을 미치지 않았지만, 아미노산 생산량은 미네랄을 처리하였을 때 증가함을 확인하였다. 본 연구는 토양에 존재하는 페놀을 유용 아미노산으로 전환시키는데 최적의 성분은 철 미네랄(FeSO4)이라는 것을 보여주었다. 또한 철 미네랄이 존재할 때, 가장 좋은 뿌리 발아율을 보이며 토양 내 독성이 감소되었음을 확인하였다. 마지막으로 실제 환경에의 적용 가능성을 알아보고자 올해 그 부산물로 인한 토양 오염의 우려로 부각되었던 매몰지와 유사한 환경을 만들어 C. glutamicum을 적용해보았다. 그 결과 C. glutamicum을 처리한 토양의 아미노산 조성이 높게 측정되었으며, 식물의 뿌리 발아율 역시 사체의 부산물로 인해 오염된 토양에서보다 향상됨을 확인 하였다. 이로써, 오염된 토양에 C. glutamicum을 적용함으로써 실제 토양 내 독성 저감 가능성을 보여주었다.

Three sequential steps were employed to accomplish this: i) identification of the effect of minerals supplementation on the cell growth and phenol degradation, ii) analysis of changes of degraded phenol and converted to various amino acids in contaminated soil environment, iii) application of ability of C. glutamicum on livestock mortalities burial. Therefore, this research may show enhanced bioavailability to produce eco-friendly technology of restoration of soil environment as a result of utilization of environmental contaminants by C. glutamicum. In this study, we conducted the analysis of phenol degradation, amino acids composition and plant-bioassay, in order to determine that whether the bioavailability of phenol is enhanced by minerals supplementation or not. First, these minerals were not affected on growth rate and efficiency of phenol degradation in C. glutamicum. Interestingly, when phenol-contaminated water added minerals, amount of amino acids in water was increased. The second step of this study indicated that Fe supplementation was shown the most optimal component to convert useful amino acids from phenol in soils. In addition, in the case of Fe supplementation, root elongation using oat was displayed less toxicity in C. glutamicum or with mineral supplementation treated sample than untreated sample. The aim of the last step is the application of C. glutamicum to restore polluted soil environment. Composition of amino acids in soil treated C. glutamicum is generally higher than that of amino acids in untreated soils. Also, root length of oat was similar to control (no diluted contaminated soil; 100% sea sand). It suggested the possibility that could be reduced toxicity of pollutants contaminated soils from grave land with disease animals.
