(2005) 분류체계와 같이 강력히 지지되는 단계통군이며 Coelogyninae 아족에 속하는 것으로 나타났다. 자란속의 계통은 B. yunnanensis 와 B. szetschuanica 두 종이 86%의 높은 bootstrap 수치로 가장 기부에 위치하였고, 자란은 B. formosana의 자매군으로 나타났다. 중국에서 채집된 미동정 자란속 식물(unknown Bletilla species)은 DNA barcoding study에 의해 B. szetschuanica로 확인되었다. 두 번째로, 자란의 자연개체군을 식물사회학적 방법으로 분류하고, 식생과 토양과의 상관관계를 밝히고자 분포서열법에 의한 분석을 실시하였다. 자란개체군은 노간주나무 우점개체군(Juniperus rigida dominant populations), 산철쭉 우점개체군(Rhododendron yedoense f. poukhanense dominant populations), 붉나무 우점개체군(Rhus javanica dominant populations), 자란 전형개체군(B. striata typical populations )으로 분류되었다. 자란개체군은 주로 한반도의 서남해안을 따라 분포하고 해발고 4-40m의 해안가 낮은 사면의 평균 41.7°의 다소 급한 경사지에 위치하고 있다. 토양의 이화학적 성분을 분석한 결과 유기물함량은 2.38-6.70%, 전질소함량 0.09-0.27%, 치환성 K 1.08-1.72cmol+/kg, 치환성 Ca 3.56-7.71cmol+/kg, 치환성 Mg 1.52-3.21cmol+/kg, 양이온치환용량 5.28-16.95cmol+/kg이며, 토양 pH는 4.60-6.01인 것으로 조사되었다. 노간주나무 우점개체군은 붉나무 우점개체군보다 해발고가 낮고 유기물함량과 전질소함량이 낮으며 경사가 급한 지역에 분포하였다. 산철쭉 우점개체군은 토양 내 양이온치환용량과 pH, 전질소, 유기물함량이 가장 높은 지역에 분포하였고, 사면방위 수치는 상대적으로 낮은 곳에 분포하였다. 붉나무 우점개체군은 양이온치환용량, 전질소, 유기물함량의 수치가 상대적으로 낮은 지역에 분포하는 것으로 나타났으며 사면방위 수치는 대체로 붉나무 우점개체군이 높은 것으로 나타났다. 세 번째로, 자란 자연 집단의 유전적 다양성과 구조를 조사하기 위하여 4개 집단 72개체에 대한 ISSR (Inter-Simple Sequence Repeats) 표지자 분석을 수행하였다. 9개의 ISSR 프라이머에서 총 126개의 재현성있는 밴드를 관찰하였으며, 종 수준에서의 유전적 다양성은 P=39.1, S=0.191 H=0.136로 낮은 유전적 다양성을 보였으며, 전체 4개의 집단 중 고흥집단과 완도집단은 비교적 높은 값으로 나타났다. AMOVA(analysis of molecular variance) 분석을 통해 전체 유전변이 중 23.5%가 집단간에 기인하는 것으로 나타났고, 나머지 76.5%는 집단내 개체간의 차이에서 기인하였다. UPGMA 에 의한 유집분석 결과, 집단의 지리적 격리 정도와 유전적 연관성이 비교적 일치하는 경향을 보여주었다. 현재 자란의 자생지는 지역개발 등으로 인하여 상당히 취약한 상태로 효율적인 보전 전략의 수립이 요구된다. 마지막으로 자란의 희귀품종인 흰꽃 변이체를 이용하여 대량증식연구를 수행하였다. 흰꽃 자란 종자의 무균발아는 수분 후 150여일된 종자를 이용하여 Hyponex 배지에 파종한 결과 약 80% 이상의 발아율을 확인할 수 있었다. 기내에서 무균발아에 의해 얻어진 소식물체를 고체배지 배양과 액체배지를 이용한 현탁배양 결과 액체배지가 배양 식물체의 생장에 효율적인 것으로 나타났다. 자란 소식물체의 바이오리액터 배양에 의한 대량 증식을 통해 크기가 작은 것(약 5 mm)은 MS 기본배지를 1/4로 줄인 조건에서 생장율이 매우 높았던 반면, 약 15 mm 정도의 소식물체는 1/2 MS 배지에서 생장이 가장 양호 한 것으로 나타났으며 증식된 배양체의 순화를 시도하였다. 또한 자란 원시구경(PLB: Protocorm Like Body) 증식에 의한 식물체 재분화 시스템을 통한 배양을 수행하였다. 그 결과 PLB 증식은 1/2 MS배지에 생장조절제 1.0 mg/L NAA와 1.0 mg/L BA를 사용하였을 때 양호하였고, 배양 절편체의 수를 3개 이상 군락접종 하였을 때 생존율이 가장 높은 것으로 나타났다. 본 연구를 통해 자란의 계통과 자란 자생지에 대한 생태학적, 집단유전학적 연구를 통하여 자란의 계통학적 위치를 파악하였으며, 현재의 자란 자생지 집단의 유전적 다양성이 낮으며 지역개발과 인위적 훼손 등으로 매우 취약한 상태인 것으로 조사되었다. 또한 자란의 보전과 원예적 상품화를 위해 흰꽃 변이체 자란의 대량증식방법과 고효율 재분화 체계를 확립하였다. 희귀식물인 자란의 포괄적인 종생물학적 연구를 통하여 현지내외 보전을 위한 효율적인 보전전략의 수립과 원예화를 위한 중요한 기초자료를 제공하였다.

Studies on the phylogenetic relationships and conservation biology of Bletilla striata were conducted. First, sequences of four chloroplast DNA regions including matK, rbcL, trnL intron and trnL-trnF spacer regions were analyzed to address phylogenetic relationships of the genus Bletilla species, and clarify systematic position of the genus Bletilla. The East Asian endemic genus Bletilla is strongly supported as a monophyletic group, and placed within the subtribe Coelogyninae in the classification systems of Van den Berg et al. (2005). In the Bletilla phylogeny, a clade belongs to that of B. yunnanensis and B. szetschuanica placed at the base with 86% of bootstrap value and the study species B. striata is the sister to B. formosana which is distributed in China. A putative Bletilla species from China which was sampled without flowers, identified as B. szetschuanica by DNA barcoding study. Secondly, phytosociological study was carried out to analyze the vegetation properties, soil characteristics and ordination of B. striata populations in South Korea. The B. striata populations were classified into Juniperus rigida dominant populations, Rhododendron yedoense f. poukhanense dominant populations, Rhus javanica dominant populations, and B. striata typical populations. B. striata is mainly distributed along the south-western coastal district in the Korean peninsula and the B. striata populations were located at an elevation of 4 to 40m. In the study sites, soil organic matter, total nitrogen, available phosphate, exchangeable potassium, exchangeable calcium, exchangeable magnesium, cation exchange capacity and soil pH were analyzed. Juniperus rigida dominant population was found in a low elevation and steep sloped area that has less percentage of organic matter and total nitrogen than Rhus javanica dominant population. Rhododendron yedoense f. poukhanense dominant population and B. striata typical population were found in a low elevation area that has less percentage of organic matter, total nitrogen and cation exchange capacity than other populations. Rhus javanica dominant population was found in relatively higher elevation and steep sloped area that has high percentage of organic matter and total nitrogen, exchangeable potassium, exchangeable calcium, exchangeable magnesium, and cation exchange capacity. Thirdly, the genetic diversity and structure of B. striata in four populations were investigated using 126 markers derived from nine ISSR primers. Overall genetic diversity at the species level (P=39.1, S=0.191 H=0.136) was very low which is substantial considering the limited distribution of populations. The population of Goheung and Wando Island had the highest level of genetic diversity. Based on analysis of molecular variance (AMOVA), a low degree of differentiation among-population was revealed (23.5%). The UPGMA dendrogram based on Nei’s genetic distance showed geographic isolation coincided with genetic correlations. Lastly, the white flower mutant, B. striata was mass produced by tissue culture methods. Germination of seeds on the Hyponex media was found to be the highest (80%) when mature capsules were collected five months after pollination. The germinated seeds of B. striata were cultured on solid and liquid Hyponex media. It was found that plantlet multiplication of white flower mutant B. striata was more efficient in liquid culture than solid culture. In bioreactor culture for mass production, small size of plantlets (about 5 mm) grew very well in 1/4 MS medium, whereas larger size of plantlets (about 15 mm) grew well only in 1/2 MS medium. There was an increase in the size of the plantlet by 6 to 8 times after 4 weeks. We also developed systems for effectively multiplying protocorm-like bodies (PLBs). The highest frequency (68.0%) of PLBs formation was obtained on 1/2 MS agar-solidified medium containing 1.0 mg/L NAA and 1.0 mg/L BA. To test their survival ratio, the ratio of PLBs formation went up as high as 95.7% when 3 populations were transplanted rather than those of one or two populations. The 1/2 MS medium without plant growth regulators was suitable for plantlet regeneration from PLBs. Through all these studies, we found the phylogenetic status of B. striata. The current status of B. striata habitats is very vulnerable with artificial damages and local development constantly threatening the species’ survival. Also genetic diversity of B. striata populations was found to be low. A mass production system and efficient regeneration system of white flower mutant B. striata for conservation and commercial were established. Consequently, a useful and important baseline information were provided for conservation and horticultural use through comprehensive species biology of rare orchid B. striata.
