PMA에 의해 활성화된 Raw264.7 세포에서 superoxide (O2-)의 생산이 Betanin에 의해서 억제되는 것을 Lucigenin chemiluminescence assay를 이용한 NADPH oxidase activity 활성을 통해 확인하였으며, PMA에 의해 활성화된 Raw264.7 murine macrophage세포의 NADPH oxidase는 PMA 자극 1시간 후에 최대의 활성을 보였다. Betanin의 NADPH oxidase activity 억제효과를 비교하기 위해 Apocynin이라 하는 NADPH oxidase activity 억제제를 사용하여 비교하였으며, Raw264.7 murine macrophage 세포는 PMA 자극 1 시간 후 정상세포에 비해 276.7%의 활성 상승 수치를 보였다. 대조군인 Apocynin의 경우 1mM/ml 에서 1uM/ml 까지 농도의존적으로 NADPH oxidase activity 억제를 보였다. Betanin은 PMA 자극 1시간 전에 처리하였으며, PMA 자극 1 시간 후 Betanin 100 ng/ml 농도에서 29.5%의 NADPH oxidase activity 억제효과를 보였다. 또한, Betanin과 dextrin을 분리한 순수 Betanin과 합성품인 iso-betanin을 이용하여 NADPH oxidase activity 억제효과를 볼 수 있었으며, 순수 betanin과 iso-betanin은 1 ug/ml 에서 최대 15.8%, 6.1%의 억제효과를 보였다. 이 결과로 보아 PMA에 의해 활성화된 Raw264.7 murine macrophage세포가 betanin에 의해 superoxide(O2-)의 생산이 감소되는 것으로 보이며, specific NADPH oxidase activity inhibitor인 Apocynin에 비해 미약하게 betanin이 superoxide(O2-)를 생산하는 NADPH oxidase에 작용하여 NADPH oxidase의 활성억제를 보이는 것으로 사료된다.

Raw264.7 cells, stimulated by PMA, was inhibited the superoxide (O2-) production by Betanin using Lucigenin chemiluminescence assay and confirmed the maximal activation level of NADPH oxidase activity at 1 hour after the PMA stimulated. The NADPH oxidase activity levels was decreased of 29.5% by betanin 100 ng/ml than PMA stimulated Raw 264.7 cells. In addition, pure betanin and iso-betanin treated group (1 ug/ml) were reduced NADPH oxidase activity levels 15.8%, 6.1% than PMA stimulated group. From these results. the activated NADPH oxidase activity in Raw264.7 murine macrophage cells were inhibited by betanin and shown that similarly effect of Apocynin, specific NADPH oxidase activity inhibitor. Betanin is widely used for purpose of antioxidant effect chemicals and numerous experimental studies suggest that it acts as a free radical scavenger. In this experimental studies, the betanin could be inhibition effect of NADPH oxidase activity, and suggest that betanin is a effective antioxidant chemical of Reactive oxygen species.
