IMS 방법으로 식품에서 바이러스를 농축하기 위한 항체는 A형 간염바이러스는 기존의 상업적으로 판매하는 단가항체를 이용하였으며, 노로바이러스의 경우, 노로바이러스 GII/4 유전자형을 baculovirus 발현시스템을 이용한 재조합 항원을 제작하였고, 이 재조합 항원을 이용하여 노로바이러스 GII/4에 대한 다가항체를 개발하여 IMS 농축방법을 위한 항체로 사용하였다. 본 연구에서 개발한 A형 간염바이러스의 real-time RT-PCR과 기존의 conventional RT-PCR 방법을 A형 간염 IgM 양성 환자의 혈청과 분변에서 비교한 결과 약 10배의 민감도 향상을 보였다. 또한 식품중에서 IMS 농축 방법의 회수율을 측정하기 위해, 각 바이러스의 양성 환자의 분변 샘플을 양상추와 깻잎에 접종한 후, threonine buffer을 이용하여 바이러스를 탈리 한 후에, 기존에 연구된 polyethylene glycol (PEG) 침전, ultrafiltration (UF) 농축 방법과 회수율을 비교하였다. A형 간염바이러스의 경우 양상추와 깻잎의 회수율이 IMS 0.65%, 0.7%, PEG 침전 5.7%, 5.6%, UF 60%, 40.8%로 각각 나타내었고, 노로바이러스의 경우 각각의 회수율이 IMS 65.5%, 62%, PEG 침전 6.2%, 5.9%, UF 62.9%, 41.3%로 나타났다. The major causes for such phenomenon are known to be norovirus (NoV) and heaptitis A virus (HAV). The detection of virus infection by food is considered a very important factor in dealing with human disease. In most cases, difficulty arises in identifying its cause due to a lack of research on finding an appropriate method that detects when food is contaminated by viruses. This study now suggests a more accurate means of detecting foodborne viruses by two processes: separating foodborne viruses more efficiently from food using Immunomagnetic separation (IMS) and by detecting the least amount of viruses using real-time RT-PCR in virus-specific base sequence. For this purpose, the real-time RT-PCR method of HAV was developed; the existing theses were used for NoV. The antibody for enriching viruses in food with IMS used monoclonal antibody that was sold commercially for HAV. In the case of NoV, recombinant antigen was produced using baculovirus expression system for NoV GII/4 genotype. A multivalent antibody on NoV GII/4 was developed using this recombinant antigen used as the antibody for IMS enrichment. Comparing the serum and the feces of Igm-positive patient with real-time RT-PCR of HAV (developed in this study and the conventional RT-PCR) resulted in an improvement of sensitivity by approximately 10 times. To measure the yield of IMS enrichment in food, the fecal samples of the patient, who tested positive for each virus, were inoculated into lettuce and sesame leaf. After desorption of the virus using threonine buffer, a comparison was made between the yield and polyethylene glycol (PEG) precipitation as well as the ultrafiltration (UF) enrichment studied previously. In the case of HAV, the yield of lettuce and sesame leaf was shown to be 0.65% and 0.7% for IMS; 5.7% and 5.6% for PEG precipitation; and 60% and 40.8% for UF, respectively. In the case of NoV, each yield was 65.5% and 62% for IMS, 6.2% and 5.9% for PEG precipitation, and 62.9% and 41.3% for UF, respectively. In the case of HAV, UF enrichment showed the highest yield. However, this is considered to be due to the low potency of the hepatitis A antibody that is sold commercially and the use of monoclonal antibody. If a high potency and multivalent antibody was used as in the case of the NoV antibody developed from this study, then IMS enrichment can be used as an effective enrichment method in contaminated food.
