본 연구에서는 나노소재의 합성과 나노소재의 복합적인 구조를 이용하여 바이오센서, 바이오탐침, 미세유체칩연구 등을 제작하고 이의 특성을 평가 하였다. 본 논문은 1) 양자점을 이용한 바이오센서 연구, 2) 비드-양자점 복합체를 이용한 미세유체칩에서의 DNA 혼성화 검출 연구, 3) 바이오이미징 연구를 위한, 나노소재의 합성, 기능화와 그 특성 연구, 4) 양자점을 광섬유센서에 적용한 형광특성 연구로 구성되어 있다. 첫째로, 양자점을 이용한 바이오센서 연구에서, 형광공명에너지전달 (FRET) 현상을 이용한 특정유전자의 혼성화를 통해 유전자를 검출하는 양자점기반의 나노바이오센서를 개발하였다. 유전자 검출을 위한 양자점기반의 나노바이오센서는labeling 없이 삽입염료만으로 hybridization을 통해 쉽게 확인 할 수가 있고, FRET을 통해 유전자의 검출농도를 알 수가 있다. 또한, 양자점과 보조인자인 NADP+사이의 전자전달(Electron Transfer, ET) 현상을 이용해 신경전달물질인 GABA를 검출할 수 있는 바이오센서를 개발하였다. 둘째로, 비드-양자점 복합체를 이용한 미세유체칩에서의 DNA 혼성화 검출 연구는 양자점과 삽입염료의 형광소광현상을 이용해서 용액상태에서 p53 유전자의 혼성화 검출 연구를 확인한 후에 PDMS 기반의 미세유체칩에서 연구하였다. 센싱 물질로써 미세채널에 충진할 비드-양자점 복합체는 아민기를 가진 폴리스틸렌 비드와 카르복실기를 가진 양자점을 커플링 반응을 통해 제작하였다. 둑구조와 기둥구조를 지닌 미세유체칩의 미세채널에 채워진 양자점의 형광은 타겟 DNA와 탐침 DNA의 혼성화 후에 삽입염료에 의해 소광되어 진다. 다양한 미세유체칩의 구조를 통해 탐침과 타겟 유전자의 혼성화에 의한 형광 소광이 특정유전자를 검출하기 위해 연구되었다. 셋째로, 바이오이미징 연구를 위한 바이오탐침에 사용가능한 나노소재의 합성, 기능화와 그 특성을 연구하였다. 우선, 졸겔법을 이용하여 코어쉘 구조의 메조포러스 실리카를 합성하여 형광염료를 결합시키고, 그 형광특성을 연구하였다. 그 다음 입자크기가 일정한 CdSe와 CdSe/ZnS 양자점을 합성하고, 이를 구형의 실리카 네트워크에 다중으로 양자점을 결합시켰다. 수용성 양자점 마이셀은 계면활성제 마이셀에서 소수성 양자점을 연결함으로써 간단하고 빠르게 합성하였다. 마이셀의 안쪽으로 실리카의 성장과 표면에 소수성기를 가진 양자점의 연결은 성공적으로 합성되었다. 결과적으로, 수용성 양자점 마이셀은 생체분자 결합에 응용되어질 수 있고, 산소의 접근과 같은 환경적인 스트레스를 막아줌으로써 화학적, 물리적인 안정도를 향상시킬 수 있다. 마지막으로, 양자점을 광섬유센서에 고정화하여 형광특성을 연구하였다. 우선, 실린지 펌프를 사용해서 다양한 형태의 침상 광섬유를 제작했고, 양자점을 광섬유 표면에 붙이기 위한 기능화를 연구하였다. 제작된 침상 광섬유는 다양한 입자크기의 CdSe/ZnS 양자점을 사용하여 광섬유 온도센서를 제작하였다. 양자점은 EDC/NHS 커플링 반응을 통해 광섬유의 표면을 실란화함으로써 침상 광섬유의 표면에 고정화 되었고, 임의의 온도에서 광섬유 표면에 고정된 양자점의 형광 강도의 상관성이 연구되어졌다. 또한, 광섬유의 표면처리와 온도센서 적용에 따른 양자점의 광학특성이 연구되어졌다. 이러한 연구들을 통해서 나노소재를 이용한 바이오센서의 제작과 미세유체칩에서 특정 DNA 탐지가 가능함을 보였다. 특히 양자점을 이용한 응용은 양자점의 형광특성을 이용하여 유기형광염료를 대체 할 수 있고, 이를 이용한 바이오헬스케어 분야에 앞으로 각광받을 수 있는 소재로써의 가능성을 보여주고 있다. 또한, 미세유체칩에서의 형광집광을 통한 바이오칩은 유전자의 증폭이나 유기염료의 라벨링 없이 손쉽게 목표 바이오분자를 검출할 수 있어 앞으로의 예방의학분야에서의 전망을 밝게 하고 있다.

In this study, a variety of nanomaterials have been synthesized and biosensor, bioplobe, and microfluidic chips were fabricated and characterized using complex structure of nanomaterials. This study consists of four main research parts; 1) biosensor using QDs, 2) microfluidic chip using QDs, 3) bioprobe for bioimaging using organic dye and QDs, and 4) optical fiber sensors using QDs. Firstly, QD-based nanobiosensors were fabricated to detect specific gene using fluorescent resonance energy transfer (FRET) phenomena. The human genomic specific gene could be identified by the hybridization of probe and target DNA through FRET between QDs and intercalating dyes without any labeling. A new detection scheme was developed for gamma-aminobutric acid (GABA), the chief inhibitory neurotransmitter in the vertebrate central nervous system, by using the electron transfer (ET) between QDs and NADP+. This study shows that the NADP+-functionalized QD probe system can be a new tool to detect low concentration of GABA. Secondly, PDMS-based microfludic chips were fabricated to detect p53 gene, a tumor repressor gene, via fluorescence quenching of QDs by an intercalating dye. By using coupling reaction between polystyrene beads with amine functional group and QDs with carboxyl group, bead-QDs complex were prepared and packed into the microchannel as a sensing media The fluorescence of QDs could be quenched by intercalating dye after hybridization of target DNA and probe DNA at the channel of microfluidic chip with a weir structure and pillar structure. Fluorescence quenching by hybridization of probe and target DNA was studied. Thirdly, a series of nanomaterials have been synthesized and functionalized to be used as a bioprobe for bioimaging. Nano sized mesoporous silica were prepared by using sol-gel reaction and fluorescent dyes were incorporated into the silica network, and their fluorescent properties were investigated. Monodispersed CdSe and CdSe/ZnS nanocrystals were synthesized and incorporated into silica network spheres. Simple and rapid synthesis of water-soluble QD micelles was performed through encapsulation of hydrophobic QDs into surfactant micelles. The growth of the silica network inside the micelles and incorporation of QDs with hydrophobic functional groups on the surfaces were successfully performed. The resulting water-soluble QD micelles can be used for bioconjugation and have superior chemical and physical stability with less environmental stresses such as oxygen access. Lastly, QD-based optical fiber sensors have been prepared and their optical properties have been characterized. Optical fiber probes were prepared by tapering of fiber tip by acid etching using a syringe pump and functionalization of optical fiber surface to immobilize QDs. Optical fiber temperature sensors were prepared which use QDs as sensing media. CdSe/ZnS QDs with different size distributions have been used. QDs have been immobilized on the surface of tapered optical fiber tips by silanization of optical fiber followed by EDC/NHS coupling reaction, and the dependence of fluorescence intensity of QDs immobilized on the surface of optical fibers on ambient temperature has been investigated. The surface modification of optical fiber and optical characteristics of QDs along with temperature sensor application have been studied. In this study, a variety of nanomaterials and QDs were synthesized and used for the applications of biosensor, bioprobe, and microfluidic chip for specific DNA sensing. In particular, QDs can substitute conventional organic fluorescence dyes and will come into the spotlight in the field of biohealthcare, and microfluidic chip will have a bright prospect in the field of preventive medicine.
