Oryzias latipes and Oryzias sinensisis, a family Adrianichthyidae, are the species of indigenous Korean ricefishes found in fresh water of Japan, China and other East Asia nations as well as Korea. Based on morphology, allozymes and karyotype, the two species have been mainly distinguished. The present study employed the two species of ricefishes collected from 55 different locations in Korea. After morphological examination of the species, a mitochondrial cytochrome oxidase subunit I (CO I) gene was analyzed to perform species identification, investigate interspecific relationships and intraspecific genetic variability and their geographical distribution. The morphology was macroscopically examined for the presence or absence of black spots on a lateral side of body. However, this examination maybe often subjective. In order to overcome this limitation, the mitochondrial COI gene sequencing was performed and found that all individuals have the gene of 679 base pairs without any deletions and 525 base pairs of the gene were shared in the two species. Phylogenetic analysis was conducted using Neighbour-joining (NJ), Maximum-likelihood (ML) and Maximum-parsimony (MP) methods in a Mega 5 software. The bootstrap tests (1000 replicates) supported that Oryzias latipes and Oryzias sinensisis are placed into two distinct species. Thus, the two species are divided based on the mitochondrial CO I sequences, in spite of their high morphological similarities. In addition, the analyses of the CO I gene first revealed that the O. sinensisis species are present also in the upstream of the Nakdong river and the Tamjin river . Moreover, the CO I gene analyses explored that two ricefishes using the same scientific name O. latipes in Korea and Japan (medeka) may be placed in distinct groups, not the same species, given that the genetic distance (K2P) between them is 16.5%.
